摘 要:[目的]对玉露初代培养阶段影响因子进行研究。[方法]以玉露花茎为外植体,对其影响因子,如灭菌方式、植物生长调节剂、光照条件、培养部位进行研究。[结果]最佳灭菌方式为75%酒精消毒30 s+0.1%Hg Cl2消毒7 min+2%Na OCl消毒4 min; NAA和6-BA都在诱导玉露花茎脱分化中有重要作用。在一定范围内,随着二者比值增大,脱分化效果越好;最佳愈伤组织诱导培养基为MS+NAA0. 1 mg/L+6-BA 1.5 mg/L+蔗糖30 g/L+琼脂6.7 g/L;暗培养比光培养愈伤组织诱导率高;花茎的中上部和上部比中下部、下部诱导效果好。[结论]该研究为进一步筛选出适宜玉露的培养条件、缩短诱导时间提供理论依据。[ Objective] To research the impact factors on Haworthia cooped at primary culture stage. [ Method ] With H. cooped scape as the explant, we researehed the influencing tactors, such as sterilization method, plant growth regulator, illumination condition and cultivation part. [ Result] The optimal sterilization method was sterilization by 75% alcohol for 30 s+0.1% HgCl2 for 7 min+2% NaOCl for 4 min. NAA and 6-BA had important function in inducing axis dedifferentiation of H. cooperi. To some extent, the effects of dedifferentiation enhanced with the increase of NAA and 6-BA ratios. The best callus inducing medium was MS+NAA 0. 1mg/L+6-BA 1.5 mg/L. Callus induction rate was higher in dark condition than in light condition. The induction effect of the upper part was better than that of the lower part of explants. [ Conclusion] This research provided theoretieal basis tor turther screening the proper cultivation condition and shorteming the indueing time of H. cooperi.
