摘 要:【目的】采用4种不同方法进行金柑叶片蛋白质提取的相关研究,摸索出一套适合金柑叶片蛋白质的提取方法,也为其他近缘植物高质量蛋白质的提取提供科学参考依据。【方法】选择‘桂金柑一号’为供试材料,分别采用Tris-HCl提取法、尿素提取法、DTT/丙酮提取法和TCA/丙酮提取法提取叶片组织蛋白质,分析比较获得的蛋白质产率、单向电泳图和双向电泳图。【结果】不同方法的提取效果各异,Tris-HCl提取法提取的蛋白质产率、单向电泳以及双向电泳结果较理想,单双向电泳图辨识度高,蛋白点数丰富,蛋白点整齐且少有纵横纹现象。【结论】Tris-HCl提取法能够兼顾产率和质量,在4种方法中综合提取效果最好,是适用于金柑叶片蛋白质提取的理想方法。【Objective】To explore a set of effective methods for protein-extracting from Kumquat leaves and provide a scientific basis for extracting high quality protein from other related plants,four methods were used to carry out the expriment.【Method】The 'Gui kumquat No.1' leaves have been choosen as the material,the leaf tissue proteins were extracted by Tris-HCl extraction method,urea extraction method,DTT/acetone extraction method and TCA/acetone extraction method respectively.The protein yield,one-way electrophoresis and two-dimensional electrophoresis figures were analyzed.【Result】Different methods had different extraction effects.The results obtained by Tris-HCl extraction method were satisfactory in protein yield,one-way electrophoresis and double-way electrophoresis,with high recognition degree of single and double electrophoresis map,abundant protein points,orderly protein points and few streaks.【Conclusion】The Tris-HCl extraction method showed good performance in both yield and quality with the best comprehensive effects,which was an ideal extraction method of protein from kumquat leaves.
