摘 要:【目的】以灰枣和新疆林业科学院新选育的3个灰枣优系(灰实2、灰实7和灰实8)为试验材料,对其枝条组织在低温胁迫下的相对电导率、可溶性糖、可溶性蛋白、丙二醛含量及游离脯氨酸等5个抗寒性生理指标展开研究,以期为灰枣耐寒品种的选育推广提供参考。【方法】于2017年1月下旬,选取长势一致的1年生休眠二次枝进行低温胁迫试验,胁迫程度分为-15、-20、-25、-30、-35、-40℃共6个梯度,持续胁迫时间为12 h,胁迫后以5℃/h上升至0℃后恒温静置12 h测定各项指标。【结果】结合Logistic方程计算得出,灰实2、灰实7、灰实8和灰枣低温半致死温度分别为-11.48、-8.82、-9.30和-10.80℃。同时,低温胁迫过程中,可溶性糖含量均呈现出先升后降的趋势,且均在-30℃时达到最大值。可溶性蛋白含量呈现"S"型的变化趋势。丙二醛含量呈现"低-高-低-高"波浪型变化趋势,灰实2和灰枣在-35℃时含量最高,达到11μmol/g以上,灰实7和灰实8在-40℃时含量最高,为10.42~10.71μmol/g。灰实7和灰实8的游离脯氨酸含量变化幅度较大,达到3.89~3.93 mg/g;灰实2和灰枣的变化幅度较小,为3.22~3.31 mg/g。整体上看,低温处理下灰枣及其3个优系的游离脯氨酸含量变化表现为先上升后下降的变化趋势。【结论】通过抗寒因子的综合隶属度计算显示,灰实2的平均隶属度最高,为0.482;灰枣次之,为0.478;灰实8为0.476;灰实7最低,为0.456。按照平均隶属度抗寒性分级法分级,灰实2、灰实7、灰实8和灰枣的综合隶属度在0.40~0.59之间,抗寒性均为Ⅲ级,中抗,其抗寒性能力大小综合排序为:灰实2>灰枣>灰实8>灰实7。【Objective】In this study,Zizyphus jujuba'Huizao'and its three excellent strains(grey experiment 2,7,8) selected by Xinjiang academy of forestry were used as tested materials.The effects of low temperature stress on the physiological cold resistance indices of the branches of Zizyphus jujuba'Huizao',including cold relative electrical conductivity,water soluble sugar,soluble protein,malonaldehyde content and free proline content were studied to provide a reference for the breeding and promotion of cold resistant varieties of Zizyphus jujuba'Huizao'.【Method】In late January 2017,one year old dormant secondary branches were selected for low temperature stress test,the degree of stress was divided into six gradients:-15,-20,-25,-30,-35,-40 ℃,the stress time was 12 hours and then rise up to0 ℃ at 5 ℃ per hour,and after 12 hours constant temperature standing,the indices were measured.【Result】Combined with the logistic equation,the semi lethal temperatures of grey experiment 2,7,8 and Zizyphus jujuba'Huizao'were-11.48,-8.82,-9.30 and-10.80 ℃.At the same time,the content of water soluble sugar showed a trend of rising at first and then decreasing,the different varieties or excellent strains jujube trees reached the maximum value at-30 ℃.The soluble protein content showed a trend of'S'type.The MDA content in the branches of different excellent strains showed a'low-high-low-high'wave trend,the content of MDA of grey experiment 2 and Zizyphus jujuba'Huizao'reached the highest at-35 ℃,with 11μmol/g,and the MDA content of grey experiment 7 and 8 reached the highest at-40 ℃,with 10.42-10.71 μmol/g.The free proline in grey experiment 7 and grey experiment 8 changed greatly,reaching 3.89-3.93 mg/g,while the variation range of grey experiment 2 and Zizyphus jujuba 'Huizao'was smaller,which was 3.22-3.31 mg/g.As a whole,the change of free proline content showed a trend of increasing first and the decreasing at low temperature.【Conclusion】Through the calculation of
