摘  要:【目的】研究3种樟属植物对皿式培养牛樟芝(Antrodia camphorata)菌丝生长的影响,为牛樟芝的人工培养提供科学依据。【方法】以实验室保存的牛樟芝为研究材料,首先在基因组水平对其进行分子鉴定,然后在皿式培养基中分别添加一系列浓度的牛樟、香樟和樟树木屑的热水提取物、乙醇提取物,比较牛樟芝菌丝的扩散直径和菌丝状态,分析3种樟属植物对牛樟芝菌丝生长的影响。【结果】(1)经鉴定本研究用的牛樟芝为模式菌株BCRC35396,GeneBank登录号为AY378094。(2)香樟的热水提取物最能促进牛樟芝菌丝的扩散,牛樟次之,樟树最弱;综合考虑菌落的生长状态,樟树的添加是最优的,牛樟次之,香樟最差。(3)3种樟属植物乙醇提取物添加至培养基后,樟树最能促进牛樟芝菌丝的扩散,牛樟和香樟作用效果类似,次于樟树;添加樟树乙醇提取物后菌落生长状态最优,牛樟次之,香樟最差。【结论】本研究认为在牛樟芝皿式培养时可以添加牛樟或樟树木屑提取物来促进牛樟芝菌丝体的生长。该研究为牛樟芝的皿式培养提供理论基础和技术支撑。【Objective】The present study aimed to explore the effects of three Cinnamomum plants on hyphae growth of plate cultured Antrodia camphorate and supplied a scientific support for A.camphorata artificial culture.【Method】Firstly,the molecular identification at genomic level of A.camphorata preserved in our laboratory was taken,and then a series of concentration of water-extracts and ethanol-extracts of Cinnamomum kanehirae,Cinnamomum camphora var.linaloolifera and Cinnamomum camphora were added to the medium individually to explore the effect on hyphae growth of plate cultured A.camphorata through analyzing the diameter and morphology of A.camphorata.【Result】(i) It was indentified that the A.camphorata used in this study was the model strain BCRC35396,and its Gene Bank login number was AY378094.(ii) The sawdust water-extract of C.camphora var.linaloolifera added to the medium promoted hyphae fast growth of A.camphorata,followed by C.kanehirae,and C.camphora was the weakest.Considering the whole status of A.camphorata,C.camphora was the best,followed by C.kanehirae,and C.camphora var.linaloolifera was the weakest.(iii) Ethanol-extracts of three Cinnamomum plants sawdust were added to the medium.C.camphora was better than C.kanehirae and C.camphora var.linaloolifera for promoting hyphae growth of A.camphorata.The morphology of A.camphorata was the best when C.camphora sawdust ethanol-extract was added to the medium,followed by C.kanehirae,and C.camphora var.linaloolifera was the weakest.【Conclusion】The sawdust extract of C.kanehirae or C.camphora could be added to medium to promote hyphae growth of plate cultured A.camphorata.This study would provide the theoretical basis and technical support for the plate culture of A.camphorata.
