摘  要:以白色金针菇为试材,以菌丝体生物量、密度、萌发时间等为考察指标,采用单因素试验及L9(34)正交实验对影响其液体摇瓶培养时的条件因素(培养基的初始pH、接种龄、接种量、摇床转速)进行优化,以实现白色金针菇摇瓶液体菌种高密度发酵。结果表明:适宜的摇瓶培养条件为培养基初始pH 6.5,接种龄102h,接种量10%,摇床转速140r·min^(-1)。在上述条件下,金针菇菌丝体生物量达到17.93 g·L^(-1),密度104.21个·mL^(-1),直径1.35mm,萌发时间8h。White Flammulina velutipes was used as materil,the optimal culture conditions(medium initial pH,inoculum age,inoculum amount,shaker speed)were studied by single factor test and L_9(3-4)orthogonal test,indexes for mycelial biomass,density,germination time,in order to achieve the high density fermentation of white Flammulina velutipes.The results showed that the optimal culture conditions were the initial pH 6.5,inoculum age 102 hours,inoculum amount 10%,the shaking speed 140 r· min^-1.Under the conditions,the mycelial biomass of Flammulina velutipes reached17.93 g·L^-1,density was 104.21· mL^-1,the diameter was 1.35 mm,the germination time was 8 hours.
