摘  要:目的:观察火麻仁油对代谢综合征(metabolic syndrome,MS)大鼠核因子-κB(NF-κB)/单核细胞趋化蛋白-1(MCP-1)/趋化因子受体-2(CCR2)信号通路及肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)表达的影响。方法:采用喂养高盐高脂高糖饲料,建立实验性MS大鼠模型,然后用火麻仁油2. 0、8. 0ml/kg和二甲双胍70mg/kg灌胃给药,每天1次,连续4周,每天观察实验大鼠一般状况。实验大鼠分别于给药后的第2周和第4周进行实验,检测大鼠血压(BP)、肥胖相关指标(体重、腹围、体长,计算Lee's指数和肥胖指数);进行血液中空腹血糖(FBG)、胰岛素(FINS)、甘油三酯(TG)、总胆固醇(TC)、游离脂肪酸(FFA)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)和TNF-α、IL-1β和IL-6含量分析;取肝组织进行组织形态学分析;实时定量PCR检测肝组织中MCP-1和CCR2的mRNA表达; Western blot检测肝组织中MCP-1、CCR2和NF-κB p65蛋白表达。结果:火麻仁油2. 0、8. 0ml/kg和二甲双胍70mg/kg可显著改善MS模型大鼠的一般状况,降低BP、体重、腹围、Lee's指数和肥胖指数,降低血液中FBG、FINS、FAA、TG、TC、LDL-D及TNF-α、IL-1β和IL-6含量,升高血液中HDL-D含量,抑制肝组织脂肪变性及炎症细胞浸润,降低肝组织中NF-κB p65和MCP-1、CCR2表达。结论:火麻仁油对大鼠MS具有较好的治疗作用,其作用机制可能与抑制MCP-1、CCR2表达、NF-κB活化及促炎因子的生成相关。Objective: To observe the effect of hemp seed oil on nuclear factor kappa-B(NF-κB)/monocyte chemoattractant protein-1(MCP-1)/CC chemokine receptor type 2(CCR2) signaling pathway and expressions of TNF-α,IL-1β,IL-6 in the cervical spondylosis rats. Methods:The experimental MS rat model was induced by feeding with high salt,high fat and high sugar diet,and then the model rats were given hemp seed oil(2. 0 and 8. 0 ml/kg) and metformin(70 mg/kg) once a day,for four weeks,the general condition of rat was observed every day.The experimental rats were tested at the second and fourth weeks after the administration,respectively. Blood pressure(BP),obesity related indicators(such as body weight,abdominal circumference,body length,Lee’s index and index calculation of obesity),serum fasting blood glucose(FBG),FINS,TG,TC,FFA,LDL-C,HDL-C and TNF-α,IL-1β,IL-6 were measured; Hepatic histopathological changes were analyzed by HE staining; Real-time PCR was applied to detect the mRNA expressions of MCP-1 and CCR2,protein expressions of MCP-1,CCR2 and NF-κBp65 in hepatic tissue were detected by western blotting. Results: Hemp seed oil(2. 0 and 8. 0 ml/kg) and metformin(70 mg/kg) might significantly improve the general condition of the MS rats,reduce body weight,BP,abdominal circumference,Lee’s index and index calculation of obesity,serum FBG,FINS,FAA,TG,TC,LDL-D and TNF-α,IL-1β,IL-6 contents,promote serum HDL-C content; depress the mRNA and protein expressions of MCP-1,CCR2 and NF-κBp65 in hepatic tissue(P 〈 0. 05,P 〈 0. 01,respectively),inhibit hepatic steatosis and inflammatory cell infiltration. Conclusion: Hemp seed oil has a better therapeutic effect on the MS rats. Its mechanism is mainly related to inhibiting the NF-κB/MCP-1/CCR2 signaling pathway and productions of TNF-α,IL-1β,IL-6.
