骨突康贴剂对家兔的长期毒性实验研究

摘　　要：目的观察骨突康贴剂对家兔的长期毒性反应,为临床用药的安全性提供试验依据。方法 48只家兔按体重随机分组4组,空白对照组、骨突康贴剂低(0. 85g生药/kg)、中(2. 73g生药/kg)和高(8. 80g生药/kg)剂量组,经家兔皮肤给药,每天给药1次,连续给药三个月及停药2周后,对其进行一般状况观察,血液学、血液生化学检查,系统尸检、主要脏器系数及组织学检查。结果各给药组家兔在给药期间及恢复期一般状态及体重增长均无显著性改变。骨突康贴剂低、中、高剂量组血浆凝血酶原时间与空白对照组比均明显延长(P <0. 05),停药二周后恢复,其它指标无明显影响。血液生化学指标、脏器指数及各脏器组织与空白对照组比均无明显变化。结论骨突康贴剂对家兔安全剂量为8. 80g生药/kg以下,为临床成人拟用量的120倍(成人按60kg计)。OBJECTIVE To observe the chronic toxicity of Gutukang patch in rabbits,providing a scientific experimental basis for clinical treatment.METHODS Forty-eight rabbits were randomly divided into 4 groups,the control group and Gutukang patch low （0.85g·kg -1 ）,medium （2.73g·kg-1 ）and high doses groups （8.80g·kg -1 ）,Each group was transdermaladministered with drugs once a day for consecutively 3 month and then stopped giving drugs for 2 weeks for observation.The general condition,matology indexes,biochemical indicators,the major organ coefficient and histological examination were measured. RESULTS In each group,there was no significant changes in general situation and body weight in the late stage of the experiment and the ecovery period.Compared with blank control group,prothrombin time of administration group was significantly prolonged （ P 〈0.05）after 3 months and was recovered to the normal level.The other hematology indexes were no significant differences.The biochemical examination,organ coefficient and histopathology were no significant differences too. CONCLUSION The safe dose of Gutukang patch given to rabbit is over 8.80g·kg -1,it is equivalently 120 times of clinical dosage（according to 60 kg body weight of adult）.
