摘 要:目的采用HPLC法同时测定不同产地厚朴叶中芦丁、阿福豆苷、金丝桃苷、异槲皮苷、槲皮苷、和厚朴酚、厚朴酚的含量。方法采用YMC-Pack-ODS-AQ色谱柱(250×4. 6mmI. D. S-5μm,12nm),以乙腈和0. 2%磷酸水溶液为流动相进行梯度洗脱,流速为1. 0mL·min^(-1),波长为280nm,柱温为30℃。结果芦丁、阿福豆苷、金丝桃苷、异槲皮苷、槲皮苷、和厚朴酚、厚朴酚的线性范围分别为5. 45~109μg·mL^(-1),2. 51~50. 2μg·mL^(-1),1. 115~22. 3μg·mL^(-1),1. 06~21. 2μg·mL^(-1),9. 36~187. 2μg·mL^(-1),2. 3~84. 8μg·mL^(-1),1. 05~21μg·mL^(-1)。不同产地厚朴中芦丁、阿福豆苷、金丝桃苷、异槲皮苷、槲皮苷、和厚朴酚、厚朴酚的含量有所差异,可能与厚朴分布区域、土壤环境及叶片采收时间等有关。结论该方法简单、快捷、准确,可以同时测定厚朴叶中芦丁、阿福豆苷、金丝桃苷、异槲皮苷、槲皮苷、和厚朴酚、厚朴酚的含量,为厚朴叶质量控制提供实验依据。OBJECTIVE To determine the content of rutin,afzelin,hyperoside,isoquercitrin,quercetin,magnolol and honokiol in Magnoliae officinalis leaves from different habitats by HPLC.METHODS YMC-Pack-ODS-AQ(250×4.6mm I.D.S-5μm,12nm) was used,and the mobile phase consisted of acetonitrile-0.2% phosphoric acid solution with gradient elution at the flow rate of 1.0mL·min -1 ,the detection wavelength was set at 280nm and the column temperature was 30℃.RESULTS The linear ranges of rutin,afzelin,hyperoside,isoquercitrin,quercetin,magnolol and honokiol were 5.45-109μg·mL -1 ,2.51-50.2μg·mL -1 ,1.115 -22.3μg·mL -1 ,1.06-21.2μg·mL -1 ,9.36-187.2μg·mL -1 ,2.3-84.8μg·mL -1 ,1.05-21 μg·mL -1 .The content ofurutin,afzelin,hyperoside,isoquercitrin,quercetin,magnolol andhonokiol in Magnoliae officinalis leaves from different habitatsare different,and may be related to the distribution area,soil environment,and leaf harvesting time. CONCLUSION The method is simple,rapid and accurate for determining simultaneously the contents of rutin,afzelin,hyperoside,isoquercitrin,uercetin,magnolol and honokiol inMagnoliae officinalis leaves,and can provide a basis for the quality control of Magnoliae officinalis leaves.
