摘  要:目的观察水通道蛋白2 (AQP2)、γ型上皮钠通道(γ-ENaC)、Na^+/H^+交换体3 (NHE3)在"阴虚水肿"证大鼠肾脏中的表达及其意义。方法尾静脉注射阿霉素复制"水肿"状态,继予优甲乐灌胃制作"阴虚水肿"证大鼠模型,观察空白组、阴虚水肿组大鼠肾脏AQP2、γ-ENaC、NHE3表达。结果尾静脉注射阿霉素2周大鼠阴囊水肿明显,第4周阴虚水肿组大鼠皮肤含水量高于空白组(P<0.05),提示水肿形成,连续优甲乐灌胃2周阴虚水肿组大鼠活动频繁,躁动不安,易激惹,肛温、TT3、TT4高于空白组(P<0.05),符合"阴虚"表现。AQP2蛋白表达量第2周最高(P<0.05),γ-ENaC、NHE3蛋白表达量第4周最高(P<0.05)。结论 AQP2、γ-ENaC、NHE3参与"阴虚水肿"证大鼠水肿的发生发展。Objective To observe the expression and significance of aquaporin2 (AQP2) , gamma type epithelial channel ( γ-ENaC) and Na+/H+ exchanger3 (NHE3) in the kidney of " yin deficiency edema" rats. Methods Injection adriamycin from thecaudal vein to replicate the model of " edema" , then to made the rat model of " yin deficiency edema" by gavage euthyrox. Theexpression of AQP2,γ-ENaC and NHE3 in the blank group and the rats of yin deficiency edema group were observed. Results 2weeks after injection of adriamycin, the scrotal edema was obvious in rats,The water content of the skin in the fourth week was higherthan that in the blank group (P 〈 0.05) , suggesting edema formation, 2 weeks after continuous gavage euthyrox, rats in the modelgroup had frequent activities, restlessness and irritability, Anal temperature, TT3 and TT4 were higher than those in blank group (P〈 0.05) , which accords with " yin deficiency" . The expression level of AQP2 protein was highest at second weeks (P 〈 0.05),and the expression level of gammaγ-ENaC and NHE3 was highest at fourth weeks (P 〈 0.05). Conclusion AQP2, gammaγ-ENaCand NHE3 participated in the development of edema in rats with " yin deficiency edema".
