摘 要:目的:探讨蝉花菌丝对AngⅡ作用下的肾小管上皮细胞Ⅰ型胶原(ColⅠ)、Ⅲ型胶原(ColⅢ)、纤连蛋白层(FN)、黏连蛋白(LN)分泌的影响,揭示蝉花菌丝防治高血压肾损害作用的机制。方法:以AngⅡ作用下的肾小管上皮细胞为研究对象,采用血清药理学研究方法,观察空白对照组、AngⅡ对照组和蝉花高、中、低剂量组的肾小管上皮细胞ColⅠ、ColⅢ、FN、LN分泌的情况,采用SPSS 20. 0统计软件进行统计分析。结果:与空白组比较,AngⅡ组肾小管上皮细胞ColⅠ、ColⅢ、FN、LN的分泌显著增加,差异具有有统计学意义(P <0. 01,P <0. 05);与AngⅡ组比较,蝉花各剂量组的肾小管上皮细胞ColⅠ、ColⅢ、FN、LN分泌均有减少,差异具有统计学意义(P <0. 01,P <0. 05);蝉花高剂量组ColⅠ分泌低于蝉花低剂量组(P <0. 05),蝉花高、中剂量组FN分泌低于蝉花低剂量组(P <0. 01),蝉花高剂量组LN分泌低于蝉花低、中剂量组(P <0. 01,P <0. 05),不同剂量的蝉花组之间存在量效相关关系。结论:蝉花菌丝防治高血压肾损害,干预肾纤维化进程,其机制可能是抑制肾小管上细胞细胞外基质的分泌进而抗肾小管间质纤维化。Objective:To study the effect of Cordyceps Sobolifera Mycelium(CSM) on the protein of ColⅠ、Col Ⅲ、FN、LN in tubular epithelial cells( TECs) when exposed to AngⅡand reveal the mechanism of Cordyceps Sobolifera Mycelium in prevention and treatment of hypertensive renaldamage. Methods:Renal tubular epithelial cells are stimulated by AngⅡas the research object,we useserum pharmacology research methods. The TECs were divided into normal control group,Ang Ⅱcontrol group,high dose CSM-treated group,middle dose CSM-treated group and low dose CSM-treated group. All statistical tests were performed with SPSS. Results:The protein of ColⅠ、ColⅢ、FN、LN were significantly up regulated in AngⅡcontrol group compared with normal control group(P〈0. 01,P〈0. 05). Treatments with LDCSM MDCSM and HDCSM down-regulated the expressions of ColⅠ、ColⅢ、FN、LN in a dose-dependent manner,all of which were significantly lower than that in AngⅡcontrol group( P〈0. 01,P〈0. 05). Treatments with HDCSM down-regulated the expressions of ColⅠ in a dose-dependent manner,which were significantly lower than that in LDCSM-treated group( P〈0. 05);Treatments with HDCSM and MDCSM down-regulated the expressions of FN in a dose-dependent manner,which were significantly lower than that in LDCSM-treated group( P〈0. 01);Treatments with HDCSM down-regulated the expressions of LN in a dose-dependent manner,which were significantly lower than that in LDCSM-treated group and MDCSM-treated group( P〈0. 01,P〈0. 05). There was a certain dose-effect relationship between different doses of Cordyceps Sobolifera Mycelium. Conclusion:Our study suggests that CSM may ameliorate renal tubulointerstitial fibrosis induced by AngⅡ,which is probably attributable to normalizing the expressions of Extracellular matrix in TECs.
