摘 要:目的:黄稔根不同极性部位对脂多糖(LPS)刺激小鼠单核巨噬白血病细胞RAW264.7细胞炎症保护作用的初步探讨。方法:用系列浓度(12.5、25、50、100、200μg/m L)的黄稔根的不同极性部位(石油醚部位、乙酸乙酯部位、正丁醇部位、水部位)处理RAW264.7细胞,MTT法检测细胞增殖活性;通过LPS(1μg/m L)诱导RAW264.7细胞建立炎症模型,采用系列浓度黄稔根不同极性部位处理炎症模型,MTT法检测黄稔根4个不同极性部位对细胞的保护作用。结果:在实验浓度范围内,黄稔根的不同极性部位对细胞无细胞毒性作用。与LPS模型组比较,浓度为50、100、200μg/m L黄稔根乙酸乙酯部位和正丁醇部位能显著改善LPS诱导小鼠巨噬细胞RAW264.7的炎症反应,具有较好的浓度依赖性。黄稔根石油醚部位与LPS模型组相比仅在200μg/m L浓度下有保护性作用。而不同浓度的黄稔根水部位未表现出保护作用。结论:初步证明了黄稔根乙酸乙酯部位及正丁醇部位对LPS诱导RAW264.7巨噬细胞炎症的保护作用。Objective: To investigate the protective effects of Medinilla septentrionalis( W. W. Sm.) H. L. Li extracts on the inflammation of mice mononuclear macrophage RAW264. 7 cells induced by lipopolysaccharide( LPS). Methods: The RAW264. 7 cells were treated with different concentrations( 12. 5、25、50、100、200 μg/m L) of M. septentrionalis extracts( petroleum ether,ethyl acetate,n-butanol and aqueous phase fractions),and cell viability was determined by MTT method. The inflammation model which was established by LPS( 1 μg/m L)-induced RAW 264. 7 cells was used to determine the protective effects of different polar fractions from M. ? septentrionalis. The protective effects were determined by MTT method. Results: The different polar fractions from M. ? septentrionalis had no cytotoxicity to RAW264. 7 cells in the experimental concentration range.Compared with model group,ethyl acetate and n-butanol fractions of M. septentrionalis extracts at the concentrations of 50、100、200 μg/m L could significantly improve the inflammation of RAW 264. 7 cells induced by LPS,and showed a well dose-dependent manner. Petroleum ether fraction of M. septentrionalis extract had the protective effect only at the concentration of 200 μg/m L compared with model group. However,aqueous phase fraction of M. septentrionalis extract showed no protective effect. Conclusion: These data suggest that ethyl acetate and n-butanol fractions of M. septentrionalis extracts had the protective effects on the inflammation of RAW264. 7 cells induced by LPS.
