摘 要:目的探讨小鼠骨髓间充质细胞(BMSCs)在动态牵张力作用下成骨分化过程中骨膜蛋白的表达及作用。方法应用多单元细胞拉伸装置对体外分离培养小鼠BMSCs施加不同长轴形变量的动态牵张力,检测成骨分化标记物及骨膜蛋白的表达变化,并予以外源性骨膜蛋白干预,探讨骨膜蛋白在牵张力下BMSCs成骨中的作用。结果不同动态牵张力均能够促进BMSCs分泌骨膜蛋白,其中10%形变量的动态牵张力不仅促进成骨的能力最强,同时促进BMSCs分泌骨膜蛋白的能力也最强,加入外源性骨膜蛋白后,15%形变量下的细胞成骨能力具有明显提高。结论适当的动态牵张力可促进MSCs分泌骨膜蛋白,外源性骨膜蛋白可以促进牵张成骨。Objective To investigate the expression of periostin in mice bone marrow mesenehymal cells (BMSCs) during osteogenic differentiation under uniaxial distraction. Methods Different mechanical tensions were applied on the mice BMSCs by multiple-unit uniaxial stretching device. After the finish of stretching, the samples were collected to detect the expression of osteogenic markers and the expression of periostin. Results The different uniaxial stretching forces promoted the secretion of periostin by BMSCs. The stretch tension of 10% was optimum to promote osteogenesis and the secreting ability of periostin by BMSCs, and exogenous periostin could promote the osteogenesis under 15% stretch tension. Conclusion The appropriate dynamic stretching could promote the secretion of periostin by MSCs, and the periostin may play a certain role during the process of distraction osteogenesis.
