摘 要:目的研究B细胞特异性莫洛氏鼠白血病病毒插入位点-1(BMI-1)、原癌基因(C-myc)和肝癌缺失基因-1(DLC-1)在宫颈癌中的表达及其意义。方法收集43例宫颈癌组织为A组,21例宫颈上皮内瘤变组织为B组,以及20例良性病变宫颈组织为C组。用免疫组化PV-9000二步法检测BMI-1、Cmyc和DLC-1蛋白的表达,用实时荧光定量聚合酶链式反应(RT-PCR)法检测BMI-1、C-myc和DLC-1 mRNA的表达,并分析其与宫颈癌临床病理因素的关系和三者表达的相关性。结果 A组、B组和C组BMI-1 mRNA的表达量分别为0. 93±0. 09,0. 29±0. 06和0. 17±0. 03,C-myc mRNA的表达量分别为0. 96±0. 08,0. 29±0. 04和0. 16±0. 03,DLC-1 mRNA的表达量分别为0. 11±0. 01,0. 83±0. 11和0. 95±0. 16,两两比较,差异均有统计学意义(均P <0. 05)。宫颈癌组织中BMI-1、C-myc和DLC-1的表达与患者的年龄、临床病理类型均无明显的相关性(均P> 0. 05),与患者的FIGO临床分期、淋巴结是否转移和分化程度均有明显的相关性(均P <0. 05)。DLC-1的蛋白表达水平与BMI-1、C-myc的蛋白表达水平均呈负相关(均P <0. 01)。结论在宫颈癌中,BMI-1和C-myc呈高表达,DLC-1呈低表达,其均与宫颈癌患者的FIGO分期、分化程度和淋巴结转移密切相关,联合检测BMI-1、C-myc和DLC-1可作为评价宫颈癌生物学行为和判断预后的参考指标。Objective To study the expression and significance of B - cell - specific Moloney leukemia virus insertion site - 1 ( BMI - 1 ) , proto - oncogene ( C -myc) and deleted in liver cancer - 1 ( DLC - 1 ) in cervical cancer. Methods A total of 43 cases of cervical cancer were collected as group A, 21 cases of cervical intraepithelial neoplasia as group B and 20 cases of benign lesions as group C. The expression of BMI- 1, C -myc and DLC -1 protein was detected by immuno- histochemical PV -9000 two step method. The expression of BMI - 1, C - myc and DLC - 1 mRNA was detected by real time polymerase chain reaction (RT- PCR), and the relationship with the clinicopathological factors of cervical cancer and the correlation between them were ana- lyzed. Results The expression levels of BMI - 1 mRNA in group A, group B and group C were respectively (0. 93 ±0.09), (0. 29 ± 0. 06 )and (0. 17 + 0. 03 ), the difference was statistically significant ( P 〈 0. 05 ). The expression levels of C - myc mRNA in group A, group B and C group were (0. 96 ±0. 08), (0. 29 ±0. 04) and (0. 16 ±0. 03), and the difference was sta- tistically significant ( P 〈 0. 05 ). The expression levels of DLC - 1 mRNA in group A, group B and group C were respectively (0. 11±0.01), (0. 83 ±0. 11) and (0.95 +0. 16), the difference was statistically significant (P 〈0.05). There was no significant difference in the expression of BMI - 1, C - myc and DLC - 1 in the tissues of cervical cancer with the age and clinicopathological types of the patients ( all P 〉 0. 05 ). There were significant differences in the clini- cal staging, lymph node metastasis and diffe-rentiation of FIGO ( all P 〈 0. 05 ). The positive expression rate of BMI - 1 and C- myc increased with the clinical stage and differentiation degree of tumor.
