摘 要:目的:探讨益母草碱对肥大心肌细胞p38MAPK及下游基因表达的影响。方法:以血管紧张素Ⅱ(AngⅡ)诱导肥大心肌细胞,在此基础上给予p38丝裂原活化蛋白激酶(p38MAPK)抑制剂(SB203580,10μmol/L)及低、中、高浓度益母草碱(5、10、20μmol/L)干预。分别以[~3H]亮氨酸掺入量、酶联免疫吸附测定(ELISA)和硝酸还原酶法检测心肌细胞蛋白质合成速率以及细胞上清B型利钠肽(BNP)、内皮素-1(ET-1)、AngII和一氧化氮(NO)含量;Real-time PCR法检测心肌细胞ET-1、p38MAPK、肌细胞增强因子2(MEF2)、β-肌球蛋白重链(β-MHC)、α-肌球蛋白重链(α-MHC) mRNA的表达水平。结果:与模型组比较,高剂量益母草碱组心肌细胞蛋白质合成速率、BNP、ET-1及AngII含量明显降低(P〈0.05),同时ET-1、p38MAPK、 MEF2、β-MHC mRNA表达水平明显下调(P〈0.05),而NO含量和α-MHC mRNA表达水平明显上调(P〈0.05)。结论:高剂量益母草碱(20μmol/L)可能通过调节p38MAPK及下游基因表达逆转心肌细胞肥大。Objective: To investigate the effects of Leonurine on p38 MAPK and downstream gene-expression in cardiomyocyte hypertrophy. Methods: The cardiomyocyte hypertrophy was induced by AngⅡ(0.1 μmol/L)in primary neonatal cardiomyocytes. The cells were treated with p38 MAPK inhibitor(SB203580, 10 μmol/L) and different concentrations of Leonurine(5 μmol/L,10 μmol/L, 20 μmol/L).The protein synthesis was assayed with [~3H]-leucine incorporation.The contents of BNP,ET-1 and AngII were detected by ELISA,and that of NO by Nitrate reductase. The mRNA expressions of ET-1,p38 MAPK,MEF2,β-MHC and α-MHC were detected by Real-time PCR.Results: The protein synthesis,the contents of BNP,ET-1 and AngII, and the mRNA expressions of ET-1, p38 MAPK,MEF2 and β-MHC were lower,but the contents of NO and the mRNA expression of α-MHC in high-dose Leonurine group were higherthan those in model group(all P〈0.05).Conclusion:Leonurine(20 μmol/L) could inhibit cardiomyocyte hypertrophy induced by AngⅡ, and the mechanism could be related to there gulating the p38 MAPK and downstream gene-expression.
