摘 要:目的:观察艾灸神阙穴对寒凝血瘀型痛经大鼠子宫香草酸受体相关蛋白(TRPV2)表达的影响,并部分阐释其效应机制。方法:将30只雌性Wistar大鼠随机分为3组:对照组、模型组、艾条灸脐组,每组10只。模型组与艾条灸脐组构建寒凝血瘀型痛经模型,艾条灸脐组造模后给予相应治疗。观察评估各组大鼠扭体反应情况,采用免疫荧光、Western Blot、Realtime-PCR技术,检测各组大鼠子宫TRPV2在蛋白水平及mRNA水平的表达。结果:模型组大鼠扭体潜伏期明显低于对照组(P〈0.01)、扭体次数明显高于对照组(P〈0.01),艾条灸脐组大鼠扭体潜伏期明显高于模型组(P〈0.01)、且与对照组无差异(P〉0.05),扭体次数明显低于模型组(P〈0.01)。模型组大鼠子宫TRPV2蛋白水平及mRNA水平表达量均明显高于对照组(P〈0.01),艾条灸脐组大鼠子宫TRPV2蛋白水平及mRNA水平表达量均明显低于模型组(P〈0.05或P〈0.01)。结论:艾灸通过下调子宫组织TRPV2蛋白及mRNA水平的病理性高表达而对寒凝血瘀型原发性痛经有效镇痛;通过调节肥大细胞生物功能而达到镇痛效应可能是其分子机制之一。Objective:To observe the umbilical moxibustion effect on transient receptor potential vanilloid 2 of cold coagulation and blood stasis type of dysmenorrhea rats' uterusand explain its effect mechanism.Methods:Rats were randomly divided into 3 groups:control group(group A),model group(group B),moxibustion group(group C).Rats in the group A and B were constructed as cold coagulationand blood stasis type of primary dysmenorrhea model.After Group C was given corresponding treatment. Immunofluorescence,Western blot and Realtime-PCR methods were used to test the transient receptor potential vanilloid 2(TRPV2) expression in rats' uterine.Results:Writhing latency in the model group rats was significantly lower than that of the control group(P〈0.01). The writhing score was significantly higher than that of the control group(P〈0.01).Writhing latency in moxibustion group rats was significantly higher than that of the model group(P〈0.01)and had no difference with that of the control group(P〈0.05). Its writhing score was significantly lower than that of the model group(P〈0.01).The TRPV2 contents of protein and mRNA of the model group rats were significantly higher than those of the control group(P〈0.01)and the moxibustion group's were significantly lower than those in the model group(P〈0.05 or P〈0.01).Conclusion:Navel moxibustion reduced high pathological expression of uterine TRPV2 of cold coagulation and blood stasis type of primary dysmenorrhea rats to achieve effective analgesia.It may be one of its molecular mechanisms to achieve analgesic effect by regulating the biological function of mast cells.
