摘 要:目的:研究中药地骨皮水提液对自发性高血压大鼠的降压作用,并探讨其降压机制。方法:将32只SHR大鼠随机分为4组,包括模型组,地骨皮水提物高、中、低剂量组(12.0、6.0、3.0 g/kg)。另选8只同源正常血压WKY大鼠作为空白对照组。每组大鼠按体质量灌胃给药,连续给药4周,每周测量各组大鼠血压。末次给药后,眼眶静脉丛采血检测血清醛固酮(ALD)、血管紧张素Ⅱ(AngⅡ)含量,实时荧光定量PCR测定内皮素-1(ET-1)、内皮素一氧化氮合酶(eNOS)、肿瘤坏死因子TNF-α等相关基因mRNA的表达水平。结果:与模型组比较,地骨皮水提液3个剂量组都可以降低大鼠血压,且高剂量组和中剂量组在给药3周和4周后下降显著;高剂量组大鼠血清AngⅡ含量显著降低,ALD含量较模型组有所降低,但不显著;地骨皮水提液高剂量组能有效下调TNF-α、ET-1 mRNA的表达,上调eNOS mRNA表达水平。结论:地骨皮水提液可以降低SHR大鼠的血压,还能降低血浆血管紧张素Ⅱ和ALD含量,同时显著抑制TNF-α、ET-1的表达,提高eNOS mRNA的表达。说明地骨皮水提液是通过作用肾素-血管紧张素-醛固酮信号通路而起到降压作用。Objective: To observe the anti-hypertensive effect of aqueous extract of Lycii Cortex in spontaneously hypertensive rats(SHR) and explore the mechanism of lowering blood pressure. Methods: The rats were divided into four groups,including model group, high, medium and low dose of Lycii Cortex groups(12.0,6.0,3.0 g/kg),normal control group. Blood pressure was measured every week. After 4 weekscontinuous administration,the levels of AngⅡand ALD in the blood serum were measured. Results: Compared with the model group,blood pressure in three dose groups of Lycii Cortex reduced,among which SBP and DBP of rats in high dose group were decreased significantly after three weeks. The contents of AngⅡand ALD in the blood serum of high dose group were lower than those in the model group. The expression levels of TNF-α mRNA and ET-1 mRNA were significantly lower in high dose group than those in the model group. The expression of eNOS mRNA was significantly higher than that in the model group. Conclusion: Aqueous extract ofLycii Cortex could effectively reduce the blood pressure of SHR rats and the levels AngⅡand ALDas well as significantly decrease the expression levels of ET-1 and TNF-α mRNA. At the same time it can increase the expression level of eNOS mRNA. It shows that the aqueous of Lycii Cortex plays a role in reducing pressure by acting on the rennin-angiotensin-aldosterone sinaling pathway.
