摘  要:[目的]确定提取芭蕉芋叶绿素的最佳溶剂,准确鉴定芭蕉芋中的叶绿素,优化叶绿素提取工艺。[方法]对比氯仿、甲醇、90%乙醇、95%乙醇、100%乙醇、90%丙酮、95%丙酮、100%丙酮的提取效果,确定最佳提取剂;通过高效液相色谱与质谱联用手段,准确鉴定叶绿素a和b;在研究单因素提取温度、提取时间、固液比对叶绿素提取率影响的基础上以芭蕉芋叶为材料,95%乙醇为提取剂,采用正交实验设计研究各因素的显著性及叶绿素提取最佳工艺。[结果]确定了95%乙醇为最佳提取剂;准确鉴定了提取液中的叶绿素a和b;优化最佳提取条件为提取温度60℃,提取时间4 h,固液比1∶20。[结论]采用95%乙醇作为提取剂,在优化后条件下得到叶绿素提取率为75. 71%。[ Objective ] The best solvent for extracting chlorophyll from Canna edulis Ker was obtained, and the chlorophyll in the extract was accurately identified, and the extraction process was optimized. [ Methods ] The extraction effects of chloroform, methanol,90% ethanol,95% ethanol, 100% ethanol,90% acetone,95% acetone, 100% acetone were compared to determine the best extract solvent, and the chlorophyll a and b were accurately identified by HPLC - MS. The effects of extraction tempera- ture, extraction time and solid -liquid extraction on the chlorophyll extraction from Canna edulis Ker were studied by single - factor tests. The significance of the three factors were analyzed and the optimized conditions were optimized by orthogonal meth- od using 95% ethanol as extract solvent. [ Results] The 95% ethanol was determined as the best extract solvent and the chloro- phyll a and b were accurately identified. The optimum extraction conditions were obtained as follows: extraction temperature 60 ℃, extraction time 4 h, and solid - liquid ratio 1: 20. [ Conclusion ] The extraction yield of chlorophyll was 75.71% under the optimum extraction conditions by using 95 % ethanol as extract solvent.
