摘 要:通过对纯钛进行喷砂酸蚀、二次阳极氧化和载银,研究其载银量对细胞毒性及牙龈卟啉单胞菌的抗菌效果。以阳极氧化作为对照组,添加不同浓度的硝酸银为试验组,采用CellCountingKit.8(CCK-8)法检测载银对MC3T3.E1成骨细胞毒性,通过扫描电镜和能谱观察表面形貌特征和元素组成.并使用激光共聚焦显微镜观察试件表面牙龈卟啉单胞菌状态并计算其活菌比例。结果表明:随电解液中硝酸银浓度的增加,试件表面的栽银量也随之增加;CCK.8的检测结果显示3d硝酸银浓度为0.04,0.05g/L和5d硝酸银浓度为0.03,0.04,0.05g/L的毒性等级均为2级,其余均为0-l级,硝酸银浓度为0.03g-L的抗菌效果最好;电解液中硝酸银浓度为0.02g/L无明显细胞毒性,且具有良好的抗菌性能。Pure titanium was treated by sandblaster etching, secondary anodic oxidation and silver loading, and the effect of silver content on the cytotoxicity and antibacterial of porphyromonas gingivalis was studied. Apoptosis of MC3T3-E1 osteoblasts was examined by Cell Counting Kit-8 (CCK-8) by using anodic oxide film as a control group and silver-load anodic oxide film as experimental groups. The morphology and element composition of the surface were observed by scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS). The status of P. gingivalis on the surface of the specimen was observed by laser confocal microscopy, and the proportion of viable cells was further calculated. Results showed that as the concentration of silver nitrate in the electrolyte increased, the amount of silver on the specimen surface also increased. The results of CCK-8 presented that when the concentration of 3 d silver nitrate was 0.04, 0.05 g/L, and 5 d silver nitrate concentration was 0.03, 0.04, 0.05 g/L, the toxicity levels were Grade 2, and the others were grade 0-1. The optimal antibacterial effect was obtained when the concentration of silver nitrate was 0.03 g/L. Furthermore, when the silver nitrate concentration was 0.02 g/L in electrolyte, the as- prepared sample had little cytotoxicity and excellent antibacterial properties.
