摘 要:为了挖掘具有亚麻脱胶潜力的微生物资源并验证其在温水沤麻中的脱胶效果,本文首先考察了蜡样芽胞杆菌(Bacillus cereus) HDYM-02产果胶质降解酶的能力。结果表明,魔芋粉和果胶能显著诱导供试菌株产生两种果胶质降解酶,其中果胶裂解酶的活力最大值分别为(1 678. 33±3. 68)和(1 602. 67±14. 01) U·mL^(-1),果胶酸裂解酶的活力最大值分别为(670. 67±5. 31)和(488. 33±4. 92) U·mL^(-1)。进而构建加菌沤麻体系,沤麻液中的果胶裂解酶和果胶酸裂解酶的活力最大值分别为(721. 86±16. 02)和(213. 99±0. 70) U·mL^(-1),显著高于不加菌的自然对照组(P <0. 05)。同时,加菌组亚麻纤维失重率达到16%,亚麻原茎中的果胶和果胶酸利用率分别为(85. 88±0. 69)%和(63. 09±2. 16)%,均显著高于自然对照组(P <0. 05),比自然对照组分别提高了1. 12和1. 85倍。结果表明,供试菌株B. cereus HDYM-02可同时产生两种果胶质降解酶,这两种酶能高效去除亚麻胶质组分,在亚麻生物脱胶领域有广阔的应用前景。In order to excavate the microbial resources with the potential of flax degumming and verify its degumming effect in warm water retting. Firstly, this study investigated the ability of Bacillus cereus HDYM-02 to produce pectinolytic enzymes. The results showed that konjac flour and pectin significantly induced the test strain to produce two pectinolytic enzymes, wherein the maximum pectin lyase activity was (1 678.33 ±3.68) U ~ mL-1 and (1 602.67 ±14.01) U·mL^-1, and the maximum pectate lyase activity was (670.67 ± 5.31 ) U·mL^-1 and (488.33 ± 4.92)U·mL^-1, respectively. Furthermore,the bacteria-adding retting system was constructed, the maximum values of pectin lyase and pectate lyase were (721.86 ± 16.02)U·mL^-1 and (213.99 ±0.70) U·mL^-1, respectively, significantly higher than those in the natural retting system( P 〈 0.05 ). At the same time, the weight loss rate of flax stem in the bacteria-adding retting system was eventually maintained at about 16%, and the pectin and pectate utilization rates were also significantly higher than those in natural retting system, reaching (85.88±0. 69) % and (63.09 ±2.16)% , which were 1.12 and 1.85 times higher than those in natural retting system, respectively. The results indicated that the test strain B. cereus HDYM-02 can simultaneously produce two pectinolytic enzymes and efficiently remove the flax gum, which has broad application prospects in the field of flax bio-degumming.
