摘 要:目的筛选马钱子致神经毒性部位和白芍神经保护作用的部位,为马钱子临床安全用药提供依据。方法灌胃给予大鼠马钱子不同提取物(水提取物、50%乙醇提取物、总生物碱),通过血清生化指标和组织病理学筛查马钱子致神经毒性作用部位;在马钱子致神经毒性作用部位基础上,灌胃给予大鼠白芍不同提取物(水提取物、70%乙醇提取物、白芍总苷),通过血清生化指标和组织病理学观察结果,确定白芍神经保护作用部位。结果总生物碱与洋金花阳性对照组的结果一致,大鼠血清生化指标与空白组相比出现显著变化,脑组织切片显示严重的脑损伤;白芍总苷预先给药组的大鼠血清生化指标结果与空白组相比无显著性差异,脑组织切片未见明显损伤。结论马钱子致神经毒性部位为马钱子总生物碱;白芍的神经保护作用部位为白芍总苷。本研究结果为马钱子安全合理用药提供了依据。Objective To look for the fraction of Semen Strychni induced neurotoxicity and the active fraction of Paeonia lactiflora Pall. exerted neuroprotective effects. To supply a guidance for the safe utilization of Semen Strychni in clinic. Methods Using different approaches to distill Semen Strychni and Paeonia lactiflora Pall. A reasonable scheme was designed for orally administrating rats and collecting bio-samples.Target fractions of Semen Strychni induced neurotoxicity and Paeonia lactiflora Pall. exerted neuroprotective effects were guided by the serum bio-chemical and histopathology methods respectively. Results The results of serum bio-chemical and histopathology methods showed that rats in the dichloromethane phase( SAE) of Semen Strychni group suffered from severe neurotoxicity and brain injury,while,the results of rats in total glycosides of paeony group was consistent with that in blank group. Conclusion The dichloromethane phase of Semen Strychni induced neurotoxicity and total glycosides of paeony could prevent this from happening and thus exert neuroprotective effect. This may help to utilize Semen Strychni safely in clinic and may also afford an approach for the precaution of drug-induced brain injury.
