摘 要:目的:通过对国产薏苡仁和泰国薏苡仁四段DNA条形码序列的比较,探索两者的亲缘关系,为其基因型上的差别提供佐证。方法:采用聚合酶链式反应(PCR)扩增两种薏苡仁的ITS2、psbA-trnH、rbcL、matK和ITS四段保守基因,并对其PCR产物进行测序和比对。结果:四段基因的PCR扩增和测序成功率均为100%,且国产薏苡仁与泰国薏苡仁的四段基因序列无位点差异。结论:国产薏苡仁与泰国薏苡仁的亲缘关系极近,或可一定程度上替代国产薏苡仁。Objective: To explore the vide the evidences of DNA differences genetic relationship between domestic and Thailand Semen Coicis and pro- through the sequence alignment of four DNA fragments barcodes. Methods: The polymerase chain reaction (PCR) was used to amplify DNA sequences of ITS2, psbA-trnH, rbcL, marK and ITS conserved gene of domestic Semen Coicis and Thailand Semen Coicis, and PCR product sequencing and align- ment were performed. Results: The success rates of PCR amplification and sequencing for ITS2, psbA-trnH, rbcL, and matK were 100%, and there was no difference on the DNA sequences sites between domestic Semen Coicis and Thailand Semen Coicis. Conclusion: There was very close genetic relationship between domestic Semen Coicis and Thailand Semen Coicis. It is probable that Thailand Semen Coicis could substitute domestic Semen Coicis in the market.
