摘  要:目的探讨王氏连朴饮治疗急性肺损伤的可能作用机制。方法将36只SD大鼠随机分成对照组、模型组、地塞米松组和王氏连朴饮大、中、小剂量组,每组6只,雌雄各半。造模前先连续灌胃5天,地塞米松组用地塞米松注射液(浓度为0. 9 mg/ml)灌胃,王氏连朴饮大、中、小剂量组分别给予浓度为1. 75、0. 87、0. 43 g/ml药液灌胃,对照组和模型组用等量蒸馏水灌胃,每日1次,每次1 ml。第6天采用平刺尾静脉注射脂多糖(0. 2 g/100 g)后静置24 h建立急性肺损伤模型。第7天检测大鼠血清中肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)含量,HE染色观察肺、肠组织病理形态学状态。结果与对照组比较,模型组大鼠血清TNF-α、IL-1β含量明显增高(P <0. 01);与模型组比较,地塞米松组和王氏连朴饮各剂量组大鼠血清TNF-α含量明显降低,地塞米松组和王氏连朴饮大、中剂量组血清IL-1β含量降低(P <0. 01)。与王氏连朴饮小剂量组比较,王氏连朴饮大剂量组大鼠血清TNF-α含量明显降低,王氏连朴饮大、中剂量组大鼠血清IL-1β含量亦降低(P <0. 01)。HE染色显示,王氏连朴饮大、中、小剂量组对急性肺损伤大鼠肺、肠组织病理学均有所改善,且以王氏连朴饮大剂量组效果更佳。结论王氏连朴饮对急性肺损伤大鼠肺、肠组织具有一定的保护作用,可能通过控制炎症反应,从而抑制其肠-肺轴转移。Objective To observe the effects and possible mechanism of Wangshi Lianpo Decoction( 王氏连朴饮)on acute lung injury. Methods A total of 36 SD rats were randomly divided into control group,model group,dexamethasone group and Wangshi Lianpo Decoction high,middle and low dose groups,with 6 rats in each group,half male and female. The rats were given intragastric administration for 5 days before modeling,and the dexamethasone group was intragastrically administered with dexamethasone injection( 0. 9 mg/ml). The concentration of the high,middle and low doses groups were given 1. 75,0. 87,0. 43 g/ml Wangshi Lianpo Decoction,and the control group as well as the model group were orally administered with the same amount of distilled water once a day,1 ml each time.On the 6 th day,a model of acute lung injury was established after tail-end injection of endotoxin( LPS) and was allowed to stand for 24 hours. On the 7 th day,the levels of tumor necrosis factor-α( TNF-α) and interleukin-1β( IL-1β) in the serum of rats were detected,and the histopathological state of lung and intestine was observed by HE staining. Results The expressions of TNF-α and IL-1β in serum of ALI model group were significantly increased( P〈 0. 01). Compared with the model group,serum TNF-α levels in the dexamethasone group and all Wangshi Lianpo Decoction groups were significantly decreased,and serum IL-1β levels were decreased in the dexamethasone group and Wangshi Lianpo Decoction high and middle dose groups( P〈 0. 01). Compared with Wangshi Lianpo Decoction lowdose group,the serum TNF-α content of Wangshi Lianpo Decoction high-dose group was significantly decreased,and the serum IL-1β content of Wangshi Lianpo Decoction high and middle dose groups was also decreased( P〈 0. 01).HE staining showed that the high,middle and low doses of Wangshi Lianpo Decoction had improved the lung and intestine tissues of ALI rats. The most remarkable effect occured in the high dose Wangshi Lianpo Decoction group.C
