摘  要:目的确定提取洋葱多糖的最佳工艺条件,并测定其含量。方法采用热水提醇沉法提取洋葱多糖,并用正交试验设计对提取工艺条件进行优化;采用苯酚-硫酸法测定洋葱多糖含量。结果正交试验3个因素对多糖提取率的影响强度为料液比>提取温度>提取时间,料液比为1∶20,提取温度为70℃,提取时间为2 h时,洋葱多糖提取率(4. 57%)最高,提取的洋葱多糖含量为64. 56%。结论采用该方法洋葱多糖提取率高且含量也很高,操作简便,容易实施,重复性好,可作为洋葱多糖的含量测定方法。Objective To optimize the conditions for the extraction of onion polysaccharides and determine its content. Methods Onion polysaccharide was extracted by hot water extraction and alcohol precipitation,and the extraction conditions were optimized by orthogonal test. The content of onion polysaccharide was determined by phenol- sulfuric acid method. Results The order of the influence of three factors on the extraction rate of onion polysaccharide was as follows: material-liquid ratio 〉 extraction temperature 〉 extraction time. When the material-liquid ratio was 1:20,the extraction temperature was 70 ℃ ,and the extraction time was 2 h,the extraction rate of onion polysaccharide was the highest(4. 57% ),and the content of onion polysaccharide was 64.56%. Conclusion The method was useful with high extraction rate and high content of onion polysaccharide,it is simple,easy to implement,and has good repeatability,which can be used for content determination of onion polysaccharide.
