摘 要:甜菜(Beta vulgaris)的M14品系在组织学、染色体遗传上表现出无融合生殖特征,使利用杂种优势获得优良种质成为可能。为了获得调控无融合生殖的候选基因,通过实验采用同源克隆及c DNA末端快速扩增(RACE)的方法,研究首次从甜菜M14品系中克隆了与生殖相关的基因BvMARB(Beta vulgaris matrix attachment regions binding protein)的全长cDNA序列。通过分析发现,其序列长度为2023 bp,开放阅读框为1 719 bp,编码572个氨基酸;蛋白分子量为63.17 KDa,等电点为9.2。同源比对表明,BvMARB与烟草MARB(GenBank No. AB059832)的相似度为67.20%,与番茄MARB(GenBank No. BT013761)的相似度为61.26%,与拟南芥MARB(GenBank No. AY096684)的相似度为57.19%。通过细胞内定位分析发现BvMARB所编码的蛋白主要存在于细胞核中。Sugar beet (Beta vulgaris) is an important sugar crop in China. Sugar beet (Beta vulgaris) line M14has an obvious characteristic of apomixes in histology and chromosome heredity. It is a kind of potential heterosisin high yield and elite quality inbred. In order to find out candidate genes for the regulation of apomixis, wecloned full-length cDNA sequence of BvMARB (Beta vulgaris matrix attachment regions binding protein) gene,which related to reproduction, and firstly used homologous clone method by Rapid Amplification of cDNA End(RACE). The full-length cDNA sequence of BvMARB was 2 023 bp, and had an open reading frame of 1 719 bpencoding a polypeptide of 572 amino acids. The pI and protein molecular weight deduced from the amino acidssequence was 9.2 and 63.17 kDa, respectively. Homologous alignment indicates that it was 67.20% homologouswith tobacco (GenBank No. AB059832), 61.26 % homologous with tomato (GenBank No. BT013761) and 57.19 %homologous with Arabdopsis (GenBank No. AY096684). Prediction of protein localization sites (version6.4)indicated that the coding protein of BvMARB gene mainly existed in nucleus.
