摘 要:的探讨复方木尼孜其颗粒(Munziq)对大鼠模型心肌缺血/再灌注损伤(MIRI)的影响及其机制。方法选择70只SD大鼠随机分为4组:正常假手术组(对照组)10只、正常缺血再灌注组(MIRI组)20只、复方木尼孜其颗粒组(Munziq组,5.06g/kg)20只、阿托伐他汀组(AT组,20mg/kg)20只,灌胃干预21天。检测各组心肌组织NIK、IKKα、pIKKα及p65表达及血清cTnT、CK-MB、LDH、MDA、IL-1β、IL-6、TNF-α、IL-10及ICAM-1含量。结果心肌HE染色中Munziq组和AT组心肌细胞出现轻度颗粒变性和空泡变性,红细胞和淋巴细胞浸润,血管扩张和充血几乎没有观察到;两组损伤心肌细胞的病理学特征均明显减轻,心肌细胞肿胀减少。cTnT、CK-MB、LDH及MDA水平变化:MIRI组的cTnT、CK-MB、LDH、MDA水平显著高于对照组(P〈0.05);与MIRI组比较,Munziq组和AT组的cTnT、CK-MB、LDH、MDA含量显著降低(P〈0.05)。NIK、IKKα、pIKKα及p65水平的变化:MIRI组的NIK、IKKα、PIKKα、p65水平显著高于对照组(P〈0.05);而Munziq组和AT组的上述蛋白含量明显低于MIRI组(P〈0.05)。IL-1β、IL-6、TNF-α、IL-10及ICAM-1水平变化:MIRI组IL-1β、IL-6、TNF-α、IL-10及ICAM-1水平均高于对照组(P〈0.05)。Munziq组、阿托伐他汀组和MIRI组比较差异均有统计学意义(P〈0.05)。结论复方木尼孜其颗粒在缺血再灌注损伤中具有心脏保护作用。这种作用可能通过抑制NF—κB信号通路来发挥作用。复方木尼孜其颗粒可能作为保护心脏围手术期心肌缺血再灌注损伤的干预药物。Objective To explore the effect of Munziqs on myocardial ischemia reperfusion injury (MIRI) rat and its mechanism. Methods Totally 70 rats were divide into 4 groups: Sham (Con) group (n= 10), normal ischemia - reperfusion ( MIRI ) group ( n= 10 ) , Munziq group ( 5.06g / kg ) ( n=20 ) , and Atorvastatin intervention (AT) group (20 mg/kg) (n=20). All the rats were given intragastric administration for 21 days. The expression of NIK, IKKα, pIKKα and p65 in myocardial tissue were determined by Western Blot, the level of cTnT, CK-MB, LDH, MDA, IL-1β, IL-6, TNF-α, IL-10 and ICAM-1 were detected by ELISA. Results HE staining in Fufang munziq group and Atorvastatin group, some myocardial cells showed slight granular degeneration and vacuolar degeneration. Pathological features of damaged myocardial cells were alleviated in both Fufang Muniziqi group and Atorvastatin group. MIRI group had significantly higher levels of cTnT, CK-MB, LDH and MDA than sham group (P〈0.05). Compared with MIRI group, the levels of cTnT, CK-MB, LDH and MDA in Fufang Muniziqi group and the Atorvastatin group were significantly lower (P〈0.05). compared with sham group, levels of NIK,IKKα, pIKKα and p65 in MIRI group were significantly higher(P〈0.05). However, levels of these proteins in Fufang Muniziqi group and Atorvastatin group were significantly decreased than those in MIRI group (P〈0.05). To analyze the changes of cytokines after Fufang Muniziqi treatment, the levels of IL-1β, IL-6, TNF-α and IL-10 were detected by ELISA. The levels of IL-1β, IL-6, TNF-α and IL-10 in MIR1 group were increased than sham group, with significant differences (P〈0.05). After treatment, the levels of these cytokines in Fufang Muniziqi group and Atorvastatin group were significantly reduced than those in MIRI group (P〈0.05).
