摘 要:目的探讨胰高血糖素样肽-1对晚期氧化蛋白产物诱导妊娠滋养细胞中活性氧表达的影响。方法体外培养早孕绒毛妊娠滋养细胞株HTR-8/SVneo,将其分为4组:正常对照组、AOPP (200 ug/m L)组、GLP-1 (0. 03 nmol/L)组、AOPP+GLP-1组。用Dihydroethidium (DHE)作为超氧化物阴离子荧光探针检测各组细胞中活性氧族(reactive oxygen species,ROS)含量,用流式细胞仪测定其荧光强度。用Hoechest 33258染色检测滋养细胞凋亡。结果 AOPP (200 ug/m L)刺激48 h后,AOPP组的ROS表达及细胞凋亡率明显高于对照组,差异有统计学意义(P <0. 001)。而在AOPP刺激剂量和时间不变的条件下,加入GLP-1 (0. 03 nmol/L),ROS的表达量及细胞凋亡率均减少,差异有统计学意义(P <0. 001);即ROS的表达量及细胞凋亡率在AOPP+GLP-1组较AOPP组减少。GLP-1 (0. 03 nmol/L)组中ROS的表达量及细胞凋亡率与对照组差异无统计学意义。结论胰高血糖素样肽-1可抑制晚期氧化蛋白产物诱导的妊娠滋养细胞内活性氧的产生,同时抑制妊娠滋养细胞凋亡。Objective To explore the effects of GLP-1 on the level of ROS expression induced by AOPP in trophoblast cell. Methods To cultivate a trophoblast cell line (HRT-8/SVneo) in vitro. The trophoblast cell were divided into 4 groups:normal control group,AOPP group (200 μg/mL),GLP-1 group (0.03 nmol/L) and AOPP+GLP-1 group.The level of ROS express in trophoblasts cell were tested by superoxide anion probe (DHE),and the ROS fluorescent intensity was detected by flow cytometry. Results Compared with control group,the expression of ROS and apoptosis in trophoblast cell significantly increased in AOPP group (200 μg/mL) for 48 h treatment ( P 〈0.001).The expression of ROS and apoptosis were inhibited,under the same dose and time of AOPP,when GLP-1 (0.03 nmol/L) was added.In another word,the expression of ROS and apoptosis were decreased in AOPP+GLP-1 group compared with AOPP group ( P 〈0.001).Compared with control group,the expression of ROS and apoptosis in trophoblast cell treated by GLP-1 at 0.03nmol/L did not show significant difference. Conclusions GLP-1 could inhibit AOPP-mediated induction of ROS production and apoptosis in trophoblast cell.
