摘  要:目的建立斑马鱼脑部与心脏出血两种动物模型,分析是否与临床的脾不统血证具有一致性,并观察健脾益气摄血止血效果,为临床推广应用提供依据。方法实验一:将野生型AB系60尾斑马鱼随机分为正常组和模型组两组,正常组常规饲养,模型组以50μmol/L辛伐他汀处理24 h。实验二:将黑色素等位基因突变型Albino品系斑马鱼分正常组、模型组(常规饲养)、A药物组、B药物组,以血流量、运动速度、出血发生率与出血改善率为观察指标,并分析相关数据。结果 (1)出血发生率:实验一中脑部出血发生率75%。实验二中心脏出血发生率为65%。(2)血流量测定:实验一、二:与正常组比较,模型组血流明显下降(P<0.001)。(3)运动速度:实验一:与正常组比较,模型组斑马鱼运动速度明显降低(P<0.001)。(4)出血改善率:单纯健脾益气方(A药物)对斑马鱼心脏出血无防治效果;健脾益气摄血方(B药物)能降低斑马鱼心脏出血发生率,提高出血改善率。结论 (1)辛伐他汀诱导的斑马鱼出血模型与脾不统血证临床表现基本一致。(2)健脾摄血方能降低辛伐他汀诱导斑马鱼心脏出血发生率,提高出血改善率。Objective To establish the models of the brain and the heart in zebrafish,analyze the consistency with the clinical syndrome of the failure of the spleen in controlling blood and observe the effects of the treatment for benefiting spleen qi,controlling blood and stopping bleeding so as to provide the evidence in clinical application. Methods Experiment No. 1: 60 wild zebrafish of the AB series were randomized into a normal group and a model group. In normal group,the zebrafish was fed conventionally. In the model group,simvastatin,50 μmol/L was used for 24 h. Experiment No.2: The zebrafish of Albino series,with melanin allele mutation were divided into a normal group,a model group( routine feeding),an agent A group and an agent B group. The blood flow,kinematic velocity,hemorrhage incidence and hemorrhage improvement rate were taken as the observation indexes. The relevant data were analyzed. Results 1. Hemorrhage rate: in the experiment No. 1,the incidence of cerebral hemorrhage was 75%. In the experiment No. 2,the incidence of cardiac hemorrhage was 65%. 2. Determination of blood flow: compared with the normal group,the blood flow was reduced obviously in the model group in the experiment No. 1 and No. 2( P〈0. 01). 3. Kinetic velocity: in the experiment No. 1,the kinetic velocity was reduced obviously in the model group as compared with the normal group( P〈0. 01). 4. The hemorrhage improvement rate: the simple application of jianpi yiqi formula( medicine A) has no effect of prevention and treatment for cardiac hemorrhage in zebrafish. The jianpi yiqi shexue formula( medicine B) reduced the incidence of cardiac hemorrhage and increased the improvement rate. Conclusion 1. The hemorrhage model induced by simvastatin in zebrafish is basically consistent with the clinical manifestation of the failure of the spleen in controlling blood. 2. The jianpi yiqi shexue formula reduces the incidence of cardiac hemorrhage induced by simvastatin and increase the improvement rate.
