摘 要:目的观察不同浓度吸氧下依托咪酯对幼鼠行为的影响。方法将21~22日龄SD大鼠随机分为6组:对照组(n=5):单次腹腔注射0. 9%氯化钠溶液10 mL/kg复合空气暴露2 h;空气+依托咪酯组(n=5):单次腹腔注射依托咪酯5 mg/kg复合空气暴露2 h;中浓度氧组(n=5):单次腹腔注射0. 9%氯化钠溶液10 mL/kg复合45%~50%浓度氧暴露2 h;中浓度氧+依托咪酯组(n=5):单次腹腔注射依托咪酯5 mg/kg复合45%~50%浓度氧暴露2 h;高浓度氧组(n=5):单次腹腔注射0. 9%氯化钠溶液10 mL/kg复合95%~100%浓度氧暴露2 h;高浓度氧+依托咪酯组(n=5):单次腹腔注射依托咪酯5 mg/kg复合95%~100%浓度氧暴露2 h。通过旷场实验、洞板实验及电迷宫实验观察空间认知、空间探索及学习记忆等认知功能变化。结果旷场和电迷宫实验中,6组比较差异无统计学意义(P> 0. 05)。洞板实验中,对照组、空气+依托咪酯组、中浓度氧组、中浓度氧+依托咪酯组、高浓度氧组、高浓度氧+依托咪酯组成绩依次为(7. 80±3. 35)次、(2. 45±2. 00)次、(10. 80±4. 44)次、(2. 60±1. 14)次、(4. 12±3. 00)次、(1. 60±1. 14)次,与对照组比较,空气+依托咪酯组减少(P <0. 05),中浓度氧组差异无统计学意义(P> 0. 05),高浓度氧组减少(P <0. 05);空气+依托咪酯组、中浓度氧+依托咪酯组、高浓度氧+依托咪酯组任意两两比较差异无统计学意义(P>0. 05);与中浓度氧组比较,中浓度氧+依托咪酯组减少,与高浓度氧组比较,空气+依托咪酯组、高浓度氧+依托咪酯组均减少但差异无统计学意义(P> 0. 05)。结论 2 h的95%~100%高浓度氧暴露可损害幼鼠空间探索能力,而不影响空间认知和学习记忆能力;依托咪酯对2 h 95%~100%浓度氧暴露下的幼鼠有空间探索能力保护效应; 2 h空气及45%~50%浓度氧暴露下,依托咪酯均仅损害幼鼠空间探索能力,2 h 95%~100%浓度氧暴露下,依托咪酯对幼鼠有潜在空间探索能力损害作用Objective To observe the effects of etomidate on the behavior of young rats under different concentration of oxygen. Methods SD rats of the age from 21 to 22 days were randomly divided into six groups,control group( Group Ⅰ),air + etomidate group( Group Ⅱ),45% ~ 50% concentration of oxygen group( Group Ⅲ),45% ~ 50%concentration of oxygen + etomidate group( Group Ⅳ),95% ~ 100% concentration of oxygen group( Group Ⅴ),and95% ~ 100% concentration of oxygen + etomidate group( Group Ⅵ). The cognitive function were tested with Open Field Test( OFT),Hole Board test and Y-maze test. Results In the OFT and Y-maze test,there was no significant difference among the six groups. In the Hole Board test,the times of rats stretch into the hole in Group Ⅰ,Ⅱ,Ⅲ,Ⅳ,Ⅴand Ⅵ were 7. 80 ± 3. 35,2. 45 ± 2. 00,10. 80 ± 4. 44,2. 60 ± 1. 14,4. 12 ± 3. 00 and 1. 60 ± 1. 14,respectively; while there were significant differences between Group Ⅰ and Ⅱ,and between Group Ⅰ and Ⅴ. Conclusion Exposure to95% ~ 100% concentration of oxygen for two hours can affect the exploration ability,but not spatial cognition or learning and memory. The protective effect of etomidate on the spatial exploration is found in rats exposed to oxygen at 95% ~100%; Etomidate only damages the space exploration ability in rats exposed to air and oxygen concentration of 45% ~50% for two hours; and damages the ability of space exploration.
