摘  要:目的建立快速区分白纹伊蚊和埃及伊蚊的PCR结合高分辨率熔解曲线(PCR-HRM)方法。方法设计针对白纹伊蚊和埃及伊蚊线粒体ATP6和Cytb基因的2对通用引物,用PCR-HRM方法区分白纹伊蚊和埃及伊蚊。结果白纹伊蚊和埃及伊蚊ATP6和Cytb基因均扩增出不同形状的曲线,蚊种区分结果与形态学鉴定结果一致。结论采用基于ATP6和Cytb基因的PCR-HRM能准确快速区分白纹伊蚊和埃及伊蚊,可作为白纹伊蚊和埃及伊蚊不完整样本的鉴定方法。Objective To establish PCR combined high-resolution melting curve(HRM) method for rapid identification of Aedes albopictus and Ae.aegypti.Methods Two pairs of universal primer were designed for ATP6 and Cytb genes of Ae.albopictus and Ae.aegypti mitochondrial genome.PCR-HRM method was used to distinguish these two mosquito species.Results The ATP6 and Cytb genes of Ae.aegypti and Ae.aegypti were amplified in different curve shapes.The differentiation were consistent with morphological identification.Conclusion PCR-HRM based on ATP6 and Cytb genes can accurately and quickly distinguish Ae.albopictus and Ae.aegypti,which can be used as a method to distinguish the incomplete samples of Ae.albopictus and Ae.aegypti.
