摘  要:目的分析长链非编码RNA P25201在糖尿病高渗状态(HHS)患者血清中的表达及临床意义。方法研究对象分为HHS组(46例)和正常对照组(46例),检测和比较2组间最高血糖、血清胰岛素、血清C肽、糖化血红蛋白、心肌标志物以及血清P25201表达水平等指标,并分析P25201与HHS的相关性。结果正常人血清P25201保持在相对较低的水平,HHS组患者血清P25201的表达水平升高(P <0. 01)。相关分析发现血清P25201的表达与最高血糖(r=0. 725,95%CI 0. 683~0. 845,P <0. 01)及糖化血红蛋白(r=0. 775,95%CI 0. 647~0. 848,P <0. 01)呈正相关关系,而与血清胰岛素水平(r=-0. 863,95%CI 0. 747~0. 902,P <0. 01)及C肽(r=-0. 792,95%CI 0. 658~0. 804,P <0. 01)呈负相关关系。ROC曲线分析长链非编码RNA(LncRNA) P25201作为预测HHS的生物标记物的特异性,结果提示曲线下面积为0. 852(95%CI. 0. 744~0. 926,P <0. 01)。结论血清P25201在HHS患者中表达显著升高,可作为HHS早期筛查的血清标记物。Objective To analyze the expression and significance of long chain non coding RNA P25201 in hyperton- ic diabetic patients. Methods The subjects were divided into HHS group and control group, 46 cases in each group. The highest blood glucose level, serum insulin, serum C peptide, glycosylated hemoglobin, myocardial markers and serum P25201 expression were detected and compared between the two groups. Analysis of correlation between P25201 and hyperosmolar state of diabetes mellitus. Results The serum levels of P25201 in normal subjects remained relatively low, while those in HHS group increased (P 〈 0.01 ). The expression of serum P25201 was positively correlated with the highest blood glucose (r =0.725, 95% C1 0. 683 - 0. 845, P 〈 0.01 ) and glycosylated hemoglobin (r = 0. 775, 95% CI 0. 647 - 0. 848, P 〈 0. 01 ). It was negatively correlated with serum insulin level (r = - 0. 863, 95% CI 0.747 ~ 0.902, P 〈 0.01 ) and C pep- tide (r = - 0. 792, 95% CI 0. 658 ~ 0. 804, P 〈 0.01 ). The specificity of long - stranded noncoding RNA (LncRNA) P25201 as a biomarker for predicting HHS was analyzed by ROC curve. The results indicated that the area under the curve was 0. 852 (95% CI 0.744 - 0.926, P 〈 0.01 ). Conclusion Serum P25201 is significantly elevated in HHS patients, and can be used as a serum marker for early screening of HHS.
