摘  要:赤霉素(GAs)与脱落酸(ABA)是块根发育过程中重要的调节因子,为了探讨两者的相互作用关系,本研究采用外源赤霉素及其抑制剂长期处理太子参,并利用ELISA法和实时荧光定量PCR法,分析经处理的太子参块根中脱落酸生物合成的动态变化。结果显示,对照组块根中ABA含量随着发育时期的变化逐渐增加,GA_3、GA_3+PBZ处理组ABA的积累表现为先上升后下降的趋势,PBZ处理组中,ABA含量呈逐渐上升的趋势。实时荧光定量PCR结果显示,GA_3处理30 d可显著降低PhZEP1、PhNCED1和PhAAO基因的表达水平,而显著上调PhZEP2、PhNCED2的表达水平;GA_3处理组中,PhCYP707A1和PhCYP707A2的表达量表现为先下调后上调的表达趋势。多效唑处理10 d时,PhNCED2和PhCYP707A2基因显著上调;处理30 d时,PhZEP1和PhAAO基因的表达水平显著下调;PBZ处理组中,PhCYP707A1的表达量呈现为先下调后上调的表达趋势。因此,外源GA_3对ABA具有重要的调控作用,与处理时间密切相关。Gibberellin (GAs) and abscisic acid (ABA) are important regulatory factors during the period of plant root growth. In this paper, in order to investigate the interaction relation between GAs and ABA, we treated long-term P. heterophylla using exogenous gibberellin and its inhibitor. The dynamic changes of ABA biosynthesis during treated roots of P. heterophylla were analyzed by ELISA and real-time fluorescent quantitative PCR method. The results showed that ABA content ofP. heterophylla roots increased gradually with its growth period in the control group. The accumulation of ABA in GA3, GA3 +PBZ treatment group showed a tendency of increasing first and then decreasing. The content of ABA in PBZ treatment group showed a trend of increasing gradually. Real-time fluorescent quantitative PCR analysis revealed that the expression levels ofPhZEP1, PhNCED1 and PhAA 0 were down-regulated significantly, while that of PhZEP2 and PhNCED2 were up-regulated apparently at GAa treated for 30 days in P. heterophylla roots. The expression level of PhCYP707A 1 and PhCYP707A 2 were down-regulated at early stage and up-regulated in GA3 treatment group. The expression level of PhNCED2 andPhCYP707A 2 were up-regulated significantly at PBZ treated for 10 days. PhZEP1 and PhAA 0 were down-regulated significantly at PBZ treated for 30 days. In addition, PhCYtVO7A 1 were down-regulated at early stage and up-regulated in PBZ treated group. Therefore, exogenous GA3 might play an important role in regulating ABA, and is closely related to the treatment time.
