亮菌多糖的单糖组成分析及含量测定方法

摘　　要：本文采用1-苯基-3-甲基-5-吡唑啉酮柱前衍生高效液相色谱法(PMP-HPLC)测定亮菌多糖的单糖组成,依据单糖组分比例配制混合标准品,采用硫酸-苯酚法测定亮菌多糖含量并考察其准确性。实验结果表明,亮菌多糖主要由Glc-Gal-Xyl-Man-Ara-Fuc构成,混合标准品在0. 02~0. 07 mg/m L范围内线性良好,硫酸-苯酚法精密度、稳定性、重复性实验的相对标准偏差均小于3. 0%。另外,采用甘露糖作为标准品,其标准曲线几乎与混合单糖标准曲线重合,可替代混合单糖标准品。该法测定多糖含量结果准确可靠,适用于亮菌多糖含量测定。This article analysed the monosaccharide compositions of Armillariella tabescens polysaccharide by the method of 1-pheny-3-methyl-5-pyrazolone（PMP）pre-column derivatization high performance liquid chromatographic（HPLC）.According to the proportion of sugars in Armillariella tabescens polysaccharide,a mixed standard solution was obtained.Polysaccharide can be determined through the standard curve by the phenol sulfuric acid method.The results showed that Armillariella tabescens polysaccharide was composed of Glc-Gal-Xyl-Man-Ara-Fuc,the linear relationship of standard mixed sugars was at the range of 0.02~0.07mg·m L^-1.The relative standard deviations of precision test,stability test and reproducibility test were less than 3.0%.The standard curve of mannose is almost coincident with the standard curve of mixed sugars,which can be substituted for mixed standard sugars.PMP-HPLC and phenol sulfuric acid method can be used to determine the Armillariella tabescens polysaccharide.
