摘  要:目的:观察8周有氧运动或抗阻运动训练对肥胖大鼠肝脏成纤维细胞生长因子21(FGF21)信号通路相关因子表达的影响,探讨不同运动方式通过FGF21信号通路对脂代谢的调节作用。方法:30只高脂饮食诱导的肥胖雄性SD大鼠随机分为安静对照组(OC组)、有氧运动组(OA组)和抗阻运动组(OR组),每组各10只。OA组大鼠进行8周的跑台训练,每周训练5天,跑台速度从第1周的15 m/min递增至第8周的25 m/min,训练时间从20 min递增至60 min。OR组大鼠采用负重爬梯法进行8周的抗阻训练,每2天训练1次,负荷逐级递增。完成最后一次训练24小时后,大鼠禁食过夜进行取材。取血清用来测试血脂、游离脂肪酸(FFA)、FGF21水平,肝脏组织样本用来测试过氧化物酶体增殖物激活受体α(PPARα)、FGF21、过氧化物酶体增殖物激活受体γ辅助活化因子1α(PGC-1α)、PGC-1β的mRNA及蛋白表达。结果:8周运动干预后,OA组和OR组大鼠的体重、血清FGF21、总胆固醇、甘油三脂、低密度脂蛋白胆固醇水平显著低于OC组(P<0.05,P<0.01)。OA组和OR组FGF21 mRNA表达量显著低于OC组(P<0.05,P<0.01),但OA组和OR组FGF21蛋白表达量却显著高于OC组(P<0.05,P<0.01)。OR组PPARαmRNA表达量显著高于OC组和OA组(P<0.01)。另外,OR组PGC-1α和PGC-1βmRNA表达量显著高于OC组及OA组(P<0.01),且OR组PGC-1β的蛋白表达量显著高于OC组(P<0.01)。结论:8周有氧运动或抗阻运动训练都提高了肥胖大鼠肝脏FGF21的蛋白表达水平,FGF21促进了脂质代谢并在一定程度上减轻了肥胖大鼠的体重,改善了血脂水平。相对于有氧运动,抗阻运动训练能够更加有效地激活FGF21信号通路下游信号因子PGC-1α和PGC-1β的表达,有助于促进FGF21生理作用的发挥及脂肪酸氧化。Objective To explore the effect of 8-week aerobic and resistance exercise on the expression of proteins related to the hepatic fibroblast growth factor 21(FGF21)signaling pathway in obese rats as well as the effect of different exercises on the lipid metabolism through the FGF21 signaling pathway. Methods Thirty high fat diet-induced obese male Sprague-Dawley rats were randomly divided into a control group(OC,n=10),an aerobic exercise group(OA,n=10),and a resistance exercise group(OR,n=10). The OA group performed an 8-week treadmill training 5 days/week,with the running speed increasing from 15 m/min at the 1 st week to 25 m/min at the 8 th week and the duration increasing from 20 to 60 min accordingly,while the OR group completed an 8-week resistance exercise program of loaded-climbing on a ladder once in every two days,with the load increasing gradually. The rats were killed after overnight fasting 24 hours after the last training session,and the blood samples and liver tissues were collected. The blood samples were measured the blood fat,serum free fat acid(FFA) and FGF21 level,while the liver tissues were detected the mRNA and protein expressions of peroxi-some proliferators-activated receptor α(PPARα),FGF21,PPARγ coactivator(PGC)-1α and PGC-1β. Results After 8-week exercise,the average body mass,serum FGF21,total cholesterol,triglyceride and low density lipoprotein cholesterin of the OA and OR groups were significantly lower than the OC group(P〈0.05,P〈0.01). The FGF21 mRNA expression of group OA and OR were significantly lower than group OC(P〈0.05,P〈0.01). However,the FGF21 protein expression of the former groups was significantly higher than the OC group(P〈0.05,P〈0.01). The PPARα mRNA expression in group OR was significantly higher than both group OC and OA(P〈0.01). Moreover,the PGC-1α and PGC-1β mRNA expression of group OR were significantly higher than the other two groups(P〈0.01),and the PGC-1β protein expression of group OR was sig
