摘  要:目的拟探讨8周有氧运动调控棕色脂肪(BAT)中VEGF通路相关蛋白以抵抗高脂膳食对机体不良影响的可能机制。方法 7周龄雄性SD大鼠50只,随机分为普通膳食组(N组,n=14,饲喂维持饲料)和高脂膳食组(H组,n=36,饲喂高脂饲料)。饲喂10周后,N组随机分为普通对照组(NC组,n=7)和普通运动组(NE组,n=7); H组随机分为高脂对照组(HC组,n=7)和高脂运动组(HE组,n=7)。NE与HE组进行8周中等强度有氧跑台运动干预。干预结束后对比各组大鼠体成分和肩胛间棕色脂肪湿重;测试血清甘油三酯和胆固醇含量; RT-qPCR检测BAT中血管内皮生长因子a(VEGFa)及其受体(KDR/Flk-1)、环氧合酶2(COX2) mRNA表达水平; Western Blot检测BAT中VEGFa、KDR/Flk-1、COX2、解偶联蛋白1 (UCP1)、肾上腺素能受体β3 (Adrβ3)和磷脂酰肌醇磷脂酶Cγ(PLCγ)蛋白表达水平。结果 (1)有氧运动可显著降低普通和高脂大鼠的体重、体脂含量和血脂水平;提高BAT含量和BAT占体脂量的百分比(BAT%);(2)有氧运动可提高VEGFa、KDR/Flk-1、COX2 mRNA的表达;(3)有氧运动可显著提高普通膳食组BAT内UCP1、COX2、Adrβ3和PLCγ蛋白水平;高脂膳食组VEGF通路相关蛋白的表达没有显著性差异,但有上调趋势。结论有氧运动可通过激活VEGF信号通路,上调BAT含量和活性,降低血脂水平,以达到降低体脂的目的。Objective The purpose of this study was to investigate the possible mechanism by which aerobic exercise for 8 weeks regulates VEGF pathway related proteins in brown adipose tissue to prevent the adverse effects of high-fat diet on the body. Methods Fifty seven-weeks-old male SD rats were divided into control group( N,n = 14,fed with normal chow)and high-fat diet group( H,n = 36,fed with high-fat chow) randomly. After 10 weeks feeding,the group N was randomly divided into normal control group( NC,n = 7) and normal exercise group( NE,n = 7); the group H was randomly divided into high-fat diet control group( HC,n = 7) and high-fat-diet exercise group( HE,n = 7). Group NE and HE was applied with aerobic exercise intervention( treadmill) for 8 weeks. After the intervention,the differences between body composition and interscapular brown adipose tissue of different groups were compared; serum total cholesterol and triglyceride were detected; Levels of VEGFa,KDR/Flk-1,COX2 mRNA were detected by RT-qPCR; The protein expression level of VEGFa,KDR/Flk-1,COX2,UCP1,Adrβ3,PLCγ were detected by Western Blot. Results( 1) Aerobic exercise may reduce the body weight,Lee mass,body fat mass and serum total cholesterol and triglyceride levels of obese rat,while increasing the percentage of brown adipose;( 2) Aerobic exercise may increase the levels of VEGFa,KDR/Flk-1,COX2 mRNA;( 3)Aerobic exercise may increase the protein expression level of VEGFa,KDR/Flk-1,COX2,UCP1,Adrβ3,PLCγ in normal chow groups,while there was no significant difference in the expression of VEGF pathway related proteins in the high-fat diet group,but there was an upward trend. Conclusion Aerobic exercise may activate VEGF pathway,thus increase the content and activity of brown adipose tissue,reduce blood lipids so as to regulate the body fat content.
