摘  要:目的观察附子不同给药剂量对大鼠肝细胞线粒体呼吸功能的影响,探讨附子对线粒体能量代谢影响机制。方法SPF级SD大鼠50只随机分为空白组、对照组、附子高、中、低剂量组,每组10只。空白组给予生理盐水1ml/100g灌胃,对照组给予左甲状腺素钠溶液100μg/100g灌胃,附子高、中、低剂量组分别给予附子水煎液1.05g(含生药)/100g、0.525g/100g、0.263g/100g灌胃。各组连续灌胃30天后,脱颈处死,取肝线粒体,测定各组线粒体呼吸功能。结果附子高、中剂量组线粒体蛋白含量显著升高;附子高、中剂量组ST3和ST4态耗氧量较空白组增高;附子高、中剂量组的RCR、P/O、OPR均显著降低。结论附子能够增加线粒体耗氧量和质子漏效应,进而增强机体能量代谢。Objective To observe effects on respiratory function by different doses of aconite and to study mechanism on promoting energy metabolism of aconite.Methods The 50 SPF level SD rats were divided randomly into five groups—the blank group,the thyroid hormone group,the high,the medium,and the low dose of aconite groups.Every group had 10 rats.Saline 1 ml/100 g was administered with the blank group,levothyroxine sodium solution 100μg/100 g was administered with the thyroid hormone group,1.05 g(crude drug)/100 g,0.525 g(crude drug)/100 g and 0.263 g(crude drug)/100 g water decoction of the aconite was separately administered with the high group,the medium group and the low group.All rats were administered once a day for30 days to be executed by cervical vertebra luxation.Mitochondria was extracted from liver tissue to determine mitochondrial oxygen consumption.Results The high,the medium and the low aconite dose groups of mitochondrial protein contents were significantly increased,oxygen consumption from the high and the medium dose group increased significantly in the statures of ST3 and ST4,RCR,P/O and OPR of the high and the medium dose groups were significantly lower.Conclusion The aconite can increase mitochondrial oxygen consumption and electron leak to enhance energy metabolism.
