摘  要:目的利用高通量测序技术分析重度寻常痤疮患者经口服米诺环素治疗后其肠道菌群的变化情况。方法以皮肤科门诊确诊的10例重度寻常痤疮患者为研究对象,按照指南推荐剂量予以米诺环素50mg口服,2次/d,连续治疗6周,分别在治疗前、停药后24h内收集患者粪便标本,提取DNA进行生物信息学分析。利用16S r RNA测序鉴定微生物的种类并进行菌群差异分析。结果治疗前后患者标本主要由拟杆菌门、厚壁菌门、变形菌门、放线菌门组成。两组肠道菌群多样性及在门、属、种水平上相对丰度差异均未发现统计学意义。结论口服指南推荐剂量米诺环素对BMI≤25的重度痤疮患者的肠道菌群未产生显著影响。Objective High throughput sequencing technology was used to analyze the changes of gut microbiome in patients with severe ache vulgaris after six weeks oral minocycline treatment. Methods Ten cases of severe ache vulgaris patients were diagnosed and treated with oral minocycline 50mg bid for six weeks. Stool samples were collected before ( P group) and after treatment ( PZ group), the bacterial DNA was extracted and subjected to 16S rRNA sequencing for the identification of microbial species,and the differences of gut microbiome were compared between P group and PZ group. Resuits Gut microbiome in the two groups mainly consisted of Bacteroidetes, Firmicutes,Proteobacteria and Actinobacteria. There were no significant differences in the diversity of gut microbiome, and no significant differences in the relative abundance of bacteria at the phylum, genus and species levels between the two groups. Conclusion Recommended dose of minocycline in ache treatment guideline has no significant effect on gut microbiome in patients with severe ache with BMI ≤ 25.
