摘  要:目的:通过观察针刺捻转补泻手法对自发性高血压大鼠(SHR)下丘脑肾素-血管紧张素系统(RAS)调控轴各组分表达的影响,探讨其调控血压的中枢机制。方法:56只雄性SHR随机等分为模型组、针刺组、捻转补法组和捻转泻法组。14只Wistar大鼠作为空白组。各针刺组分别采用相应的手法干预太冲穴,空白组和模型组不做针刺处理。实验第14天采用HE染色法观察下丘脑组织病理学改变,采用ELISA法和RT-qPCR技术检测下丘脑RAS调控轴各组分的表达。结果:与模型组比较,捻转补法组和捻转泻法组均能明显降低SHR血压(P<0.01),各针刺组RAS升压轴各组分表达量显著减少(P<0.01),RAS降压轴各组分表达量显著升高(P<0.01);且捻转泻法组降低血压和良性调控RAS效果较捻转补法组更明显(P<0.05,P<0.01)。结论:针刺捻转补泻手法可通过良性调节SHR下丘脑RAS各组分表达,发挥其降压作用以及对下丘脑神经元的保护效应,这可能是捻转补泻手法降压的中枢机制之一。Objective: To investigate the effects of twirling reinforcing-reducing manipulation(TRRM) on the expression of hypothalamic rennin-angiotensin system(RAS) axis components in spontaneously hypertensive rats(SHRs), and to explore the central mechanism of TTRM on the regulation of blood pressure(BP). Methods: Fifty-six male SHRs were randomly and equally divided into model group(M), single-needle acupuncture group(Z), twirling reinforcing group(B) and twirling reducing(X) group. Fourteen Wistar rats were assigned to the control group(K). The according acupuncture interventions at Taichong(LR3) acupoint were administered in the Z, B and X groups. The rats in the K and M groups received no treatment. On the 14 th day, the pathological changes of hypothalamus were observed by HE staining, and the expression of hypothalamic RAS axis components were detected by ELISA and RT-PCR. Results: Compared with the M group, the BP showed a significant decrease in the Z, B and X groups(P0.01), and the expression of the elevating-BP axis components in RAS were significantly decreased(P0.01), while the expression of the lowering-BP axis components in RAS were significantly increased(P0.01). Furthermore, the X group showed the better effects of lowering the BP and benignantly regulating the RAS than that of other groups(P0.01). Conclusion: The TTRM could significantly lower the BP and protect the hypothalamic neurons by benignantly regulating the expression of hypothalamic RAS axis components of SHRs, which may be one of the central mechanisms of TTRM on antihypertensive effects.
