摘 要:目的:探究低、中、高3种剂量抗纤灵方对阿霉素肾纤维化大鼠Ⅰ型胶原基因表达的影响。方法:60只SD大鼠随机分为假手术组,模型组,科素亚组,抗纤灵低、中、高剂量组。右肾摘除加重复注射阿霉素建立模型,治疗8周后处死,检测血肌酐、尿素氮、24h尿蛋白定量观察肾功能,Masson染色观察肾组织病变,Realtime PCR检测Col1α1、Col1α2 mRNA表达水平。结果:与假手术组比较,模型组Scr、BUN、24h-UTP显著升高(P<0.05),肾间质纤维化严重,肾间质病变平均积分显著升高(P<0.01),肾组织Col1α1、Col1α2 mRNA的表达显著升高(P<0.01)。与模型组比较,科素亚组和抗纤灵高剂量组Scr、BUN、24h-UTP显著降低(P<0.01),肾纤维化程度减轻,肾间质病变平均积分显著降低(P<0.01),Col1α1、Col1α2 mRNA表达显著降低(P<0.05,P<0.01)。与科素亚组比较,抗纤灵高剂量组BUN显著降低(P<0.05)。结论:抗纤灵方通过抑制Ⅰ型胶原基因的表达从而改善肾纤维化。Objective: To investigate the effects of three doses of low-, medium-and high-dose Kangxianling Decoction on the expression of type Ⅰ collagen in rats with doxorubicin. Methods: Sixty Sprague-Dawley rats were randomly divided into sham operation group, model group, low-, medium-, high-dose Kangxianling Decoction group, and Cossin group. The right kidney was removed and rats were injected with doxorubicin repeatedly to establish the model. Eight weeks later, the rats were sacrificed. Serum creatinine, urea nitrogen, and 24-hour urinary protein were measured for renal function. Masson staining was used to observe the renal pathological changes. Real-time PCR was used to detect the expression of Col1α1 and Col1α2 mRNA level. Results: Compared with the sham operation group, the Scr, BUN and 24 h-UTP in the model group were significantly increased(P0.05), the renal interstitial fibrosis was severe, mean score of renal interstitial lesions was significantly increased(P0.01), and the expression of Col1α1 and Col1α2 mRNA in the kidney tissue were increased(P0.01). Compared with the model group, Scr, BUN and 24 h-UTP in the anti-fibrin group and the high-dose group were significantly decreased(P0.01), the degree of renal fibrosis was reduced, mean score of renal interstitial lesions were significantly decreased(P0.01), and the expressions of Col1α1 and Col1α2 mRNA were decreased(P0.05, P0.01). Compared with the Kossuya group, anti-fibrin BUN was significantly lower in the high dose group(P0.05). Conclusion: Kangxianling Decoction can improve renal fibrosis by inhibiting the expression of type Ⅰ collagen gene.
