摘  要:目的:观察中药复方脊髓康对自然混杂体系中小胶质细胞吞噬神经元碎片的影响。方法:制备脊髓康含药血清;分离、培养原代神经细胞,建立混杂细胞体系;以谷氨酸诱导混杂体系中神经元损伤,以脊髓康含药血清干预混杂体系;免疫荧光染色法标记神经元骨架、碎片以及小胶质细胞膜蛋白,观察脊髓康对小胶质细胞吞噬功能的影响。结果:脊髓康中、高剂量组吞噬百分率及吞噬指数均高于空白组(P<0.01)。结论:脊髓康可能通过促进小胶质细胞吞噬神经元碎片作用,改善神经元生长微环境,进而促进存活神经元的修复再生。Objective: To observe the effect of TCM compound Jisuikang on microglia phagocytosis of neuronal debris in mixed-culture system. Methods: Preparing Jisuikang drug serum, isolating and culturing the primary nerve cells, establishing mixed nerve cells system. The neuron damage was induced by glutamic acid in the mixed-culture system, and was disposed by Jisuikang drug serum. Skeleton and debris of neurons and membrane protein of microglia were marked by immunofluorescent staining. Then the microglia phagocytosis of neurons fragments was observed after the intervention of Jisuikang. Results: Both of middle dose and high dose of Jisuikang group showed higher phagocytic index and phagocytic percentage than that of control group(P0.01). Conclusion: Traditional Chinese medicine compound Jisuikang may promote the repair and regeneration of surviving neurons by enhancing microglia phagocytosis of neuron debris and improving the growth microenvironment of neurons.
