摘 要:目的:研究望江南子不同溶剂提取物体外抗氧化作用及对大鼠脑缺血性损伤的作用,筛选望江南子体内外抗氧化作用的有效部位。方法:通过乙醇回流提取、不同极性溶剂萃取得到望江南子不同溶剂提取物,DPPH法和邻二氮菲-二价铁氧化法测定其抗氧化活性;采用MCAO制备SD大鼠脑缺血再灌注模型,观测提取物对大鼠的神经功能症状、脑梗死体积、脑组织MDA和SOD的影响。结果:不同溶剂提取物对DPPH自由基清除率顺序:正丁醇提取物>乙酸乙酯提取物>总提取物>氯仿提取物>石油醚提取物>萃余水相;对羟基自由基清除率顺序:乙酸乙酯提取物>氯仿提取物>正丁醇提取物>总提取物;与模型组比较,各给药组均可显著减少脑梗死体积百分比(P<0.01),修复脑缺血再灌注损伤大鼠神经功能、提高脑组织SOD活性(P<0.05,P<0.01),降低MDA含量(P<0.05,P<0.01)。结论:望江南子不同溶剂提取物均具有体外抗氧化及修复脑缺血再灌注损伤作用,其中以乙酸乙酯和正丁醇萃取物作用更强,可能和其中的蒽醌苷类成分有关。Objective: To study the antioxidant activity effect of different solvent extracts of Senmen occidentalis in vitro and its effect on cerebral ischemic injury in rats, and screen the effective fraction of antioxidant activity in vivo and in vitro. Methods: Different solvent extracts of Senmen occidentalis were obtained through ethanol reflux extraction and different polar solvents. The antioxidant activity was determined by DPPH method and orthophenanthroline-divalent iron oxidation. Focal cerebral ischemia/reperfusion model were established with middle cerebral artery occlusion with thread, and the effects of the extract on the neurological deficits, cerebral infarction volume, MDA content and SOD activity in brain tissue were observed.Results: The order of clearance on DPPH free radical of different solvent extracts was: n-butanol extract ethyl acetate extract total extract chloroform extract petroleum ether extract extraction raffinate. The order of clearance on hydroxyl radical was: ethyl acetate extract chloroform extract n-butanol extract total extract. Compared with the model group, all treatment groups could significantly reduce the volume percentage of cerebral infarction(P0.01), repair neurological deficits and improve SOD activity(P0.05, P0.01) and decrease MDA content(P0.05, P0.01) in brain tissue of cerebral ischemia/reperfusion rats. Conclusion: Different solvent extracts of Senmen occidentalis all have the effects of antioxidant activity and repairing cerebral ischemia/reperfusion injury in vitro, among which the effects of ethyl acetate extract and n-butanol extract are stronger, which may be related to the anthraquinone glycoside.
