摘 要:目的:观察电针通过SCF/c-kit通路对功能性消化不良模型大鼠胃窦组织中SCF、c-kit、Cx43表达的影响。方法:40只SPF级雄性SD大鼠随机分为空白组、模型组、阻断剂组和电针组。除空白组外均采用多因素干预法造模,造模成功后对阻断剂组大鼠和电针组大鼠进行电针治疗14d,结束后进行胃窦组织解剖取材,Western blot法测定大鼠胃窦组织中SCF、c-kit、Cx43蛋白的表达。结果:与空白组比较,模型组大鼠SCF、c-kit、Cx43蛋白表达明显降低(P<0.05),阻断剂组c-kit、Cx43表达明显降低(P<0.01);与模型组比较,电针组大鼠SCF、c-kit、Cx43表达明显增多(P<0.01,P<0.05)。结论:电针能够显著地增加胃窦组织中SCF、c-kit、Cx43的表达水平,作用于SCF/c-kit通路,有效改善胃肠动力障碍,从而影响胃肠运动,这可能是电针治疗FD胃肠动力障碍的作用机制。Objective: To observe the effects of electroacupuncture(EA) on the expression of SCF, c-kit and Cx43 in the sinuses ventriculi of rats with functional dyspepsia model gastric through the SCF/c-kit pathway. Methods: Forty SPF male SD rats were randomly divided into four groups: blank group, model group, blocker group, and EA group. Except for the blank group, the other groups were modeled by multifactor intervention method. After successful modeling, the blocker group and EA group were treated with electroacupuncture for 14 days, and the expressions of SCF, c-kit and Cx43 were measured by Western blot after the anatomy of the sinuses ventriculi. Results: Compared with the blank group, the expressions of SCF, c-kit and Cx43 were significantly decreased in the model group(P0.05), the expressions of c-kit, Cx43 were significantly decreased in the blocker group(P0.01). Compared with the model group, the expressions of SCF, c-kit and Cx43 were significantly increased in the EA group(P0.01, P0.05). Conclusion: Electroacupuncture treatment can significantly increase the expressions of SCF, c-kit and Cx43 in sinuses ventriculi and effectively improve the dyskinesis of gastrointestinal. It is indicated that the SCF/c-kit pathway is crucial for EA. It could be the mechanism of electroacupuncture for the treatment of FD gastrointestinal dysmotility.
