摘 要:目的:观察柴胡疏肝汤(CHSGD)对脂多糖(LPS)诱导的抑郁小鼠模型色氨酸(TRY)代谢通路的影响。方法:56只昆明小鼠随机分为对照组,模型组,米诺环素(MINO)组,1-甲基色氨酸(1-MT)组,CHSGD低、中、高剂量组(2.5、5、10g/kg),采用LPS造模。造模前72h,予药物治疗直至实验结束,观察小鼠造模后24h内的体质量变化,6h自由活动测试、强迫游泳试验(FST)和悬尾试验(TST)中抑郁行为学表现,检测脑组织白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和吲哚2,3双加氧酶(IDO)mRNA表达水平,五羟吲哚乙酸(5-HIAA)/5-羟色胺(5-HT)、5-HT/TRY及犬尿素(KYN)/TRY比值。结果:LPS诱导的抑郁小鼠模型出现24h内的体质量减轻,6h自由活动测试中活动计数减少,FST和TST中不动时间增加,脑组织IL-1β、TNF-α和IDO mRNA表达水平增加,5-HT/TRY比值下降,5-HIAA/5-HT和KYN/TRY比值升高(P<0.01)。CHSGD中、高剂量增加小鼠24h内的体质量和6h自由活动测试中活动计数,减少FST和TST中不动时间,抑制脑组织IL-1β、TNF-α和IDO mRNA表达,下调KYN/TRY比值,增加5-HT/TRY比值(P<0.05,P<0.01)。结论:CHSGD通过抑制脑IL-1β、TNF-α水平,进一步抑制IDO mRNA表达,正常化TRY代谢通路,改善LPS诱导的小鼠抑郁样行为。Objective: To investigate the effects of Chaihu Shugan Decoction(CHSGD) on tryptophan(TRY) metabolic pathway in acute depressive mouse induced by lipopolysaccharide(LPS). Methods: Fifty-six mouse were divided into seven groups randomly, including control group, model group, minocycline group, 1-methyltryptophan(1-MT) group, CHSGD high, medium and low dose groups(2.5, 5, 10 g/kg). Model mouse were treated by LPS. 72 h before LPS injection, mouse were received minocycline, 1-MT and CHSGD treatment until experiment finished. Weight changes in 24 h, performance in locomotor activity(LMA), forced swim test(FST) and tail suspension test(TST) were recorded. Brain IL-1β, TNF-α and indoleamine 2,3-dioxygenase(IDO) mRNA expression, 5-hydroxyindoleacetic acid(5-HIAA)/5-hydroxytryptamine(5-HT), 5-HT/TRY and kynurenine(KYN)/TRY ratios were caculated. Results: In model group, mouse weight in 24 h, activity counts in LMA were decreased, 'immobility' time in FST and TST were increased; brain IL-1β, TNF-α and IDO mRNA expression were increased; 5-HT/TRY ratios were decreased and KYN/TRY ratio was upregulated(P0.01). CHSGD(5, 10 g/kg) upregulated mouse weight and activity counts in locomotor activity, lowered 'immobility' time in FST and TST, inhibited brain IL-1β, TNF-α and IDO mRNA expression, down-regulates the KYN/TR ratio, up-regulates 5-HT/TRY ratio(P0.01, P0.05). Conclusion: CHSGD treats acute depression through down regulating brain IL-1β, TNF-α, IDO expression, normalizing TRY metabolic pathway and improving depressive behavior.
