摘 要:为确诊福建省某规模肉鸽养殖场疑似鸽痘疫情,采集病鸽皮肤痘痂组织,经处理后接种11日龄SPF鸡胚,培养7d后观察到鸡胚尿囊膜增厚,出现典型的白色痘斑。组织学观察可见皮肤表皮增生,形成厚的网状结构,上皮细胞呈不同程度的空泡变性,胞浆内有嗜酸性包涵体,呈红色、圆形或椭圆形;肺脏淤血,肺泡破裂及增殖性支气管炎。待检样本进行鸽痘病毒P4b基因PCR检测,得到特异性条带;根据P4b基因核酸序列构建系统发育树,结果显示分离株与来自美国(KC017965)和南非(KJ801920)的2株鸽痘病毒聚在一起,同源性为100%,与企鹅痘病毒、鹌鹑痘病毒和鸡痘病毒的同源性分别为99.6%,99.6%和91.1%;与金丝雀痘病毒和火鸡痘病毒同源性最低,为73.3%~75.0%。结果表明,本次暴发为鸽痘病毒感染所致,为鸽痘防控及致病机理研究奠定了理论基础。To diagnosis a suspected pigeonpox outbreak occurred in a scale of pigeon farm in Fujian province, pathogenic identification and histopathological observation were performed in this study. Thickening of allantois membrane and typical white acne marks were observed on 7 days after dif- ferent disposed. Samples of the pox-like skin scabs of sick pigeons were inoculated to 11 days old SPF chicken embryos. Histopathological features were observed including skin epidermal hyperpla- sia, thick reticular structure, varying degrees of vacuolar degeneration in the epithelial cells, and eo- sinophilic inclusion bodies in the cytoplasm,which were red,round or elliptical and gores,alveolar rupture and the proliferation bronchiolitis in the lung. Specific bands of P4b gene of pigeonpox vi- rus were observed. Phylogenetic analysis of P4b gene DNA sequences showed pigeonpox isolate in this study clustered with an American isolate (KC017965) and a South Africa isolate (KJ801920) together with 100% identity,and shared 99.6%,99.6% and 91.1% identity with the penguinpox virus,quailpox virus and chickenpox virus respectively,and shared low identities with canarypox virus and turkey pox virus (73.3%-75.0%). The results confirmed that the cause of this outbreak was pigeonpox virus,which lays a theoretical foundation for the prevention and control of pigeon- pox and research on pathogenic mechanism.
