摘  要:为了观察成纤维细胞生长因子21(FGF21)对高脂肪饲料诱导的非酒精性脂肪肝小鼠肝脏肿瘤坏死因子(TNF-α)及白细胞介素1β(IL-1β)的影响。本试验采用高脂肪饲料喂养昆明小鼠30d,建立非酒精性脂肪肝模型。将小鼠分为4组,A:模型+FGF21组;B:模型组;C:正常+FGF21组;D:正常组。A和C组分别连续给药6d,B和D两组注射等量的生理盐水,给药后眼球采血,采集血液样品,断颈处死小鼠,分离肝脏和脾脏组织,通过组织切片染色、血清生化及ELISA检测,观察FGF21对肝脏组织病理学、血清中TNF-α及IL-1β的影响。结果显示非酒精性脂肪肝小鼠经过FGF21治疗后,血清中TG、TC、NEFA、LDL-c含量显著降低,ELISA结果显示血清中TNF-α、IL-1β的表达与模型组比较都显著降低(P〈0.05)。结果表明FGF21明显改善高脂肪饲料诱导的非酒精性脂肪肝的病理变化。同时,降低脂肪肝组织脂肪变形引起的血液中TNF-α、IL-1β的表达,表明FGF21对非酒精性脂肪肝小鼠有潜在的治疗作用。To investigate the effects of fibroblast growth factor 21(FGF21)on tumor necrosis factor alpha (TNF-a)and interleukin 1 beta(IL-1β)of mice with nonalcoholic fatty liver disease induced by high fat diet. Kunming mice were fed with high fat diet for 30 d to establish of nonalcoholic fatty liver model. The mice were divided into 4 groups, A : model+ FGF21 group ; B : model group ; C : nor- mal+FGF21 group;D: normal group. A group and C group were administered continuously with FGF21 for 6 d. B and D group were both injected equal saline water. Blood was gained from the eyeballs of mice after administration. Then, the mice were decapitated, liver and spleen were saved. The effect of FGF21 on liver pathology,serum levels of TNF-α and IL-113 were observed by histo- logical staining, serum biochemical and ELISA detection. The levels of serum TG, TC, NEFA and LDL-c in nonalcoholic fatty mice were decreased significantly after FGF21 treatment. The results of ELISA kit showed that the expression of TNF-α and IL-1β in serum were significantly decreased compared with the model group(P〈0. 05). FGF21 can significantly improved the pathological changes of nonalcoholic fatty liver induced by high fat diet,reduced the expression of TNF-α,IL1β in the blood caused by fat deformation in the fatty liver tissue. The results indicated that FGF21 has a potential therapeutic effect on mice with nonalcoholic fatty liver.
