摘  要:[目的]对比研究"上火"人群与健康人群的肠道菌群种类,揭示"上火"对肠道微生物的影响。[方法]收集"上火"和健康人群的粪便,利用Illumina MiSeq高通量测序技术对粪便样本DNA测序,利用QIIME软件处理测序数据并计算Alpha多样性,利用PCoA和LEfSe分析方法挖掘两类人群的差异肠道菌群种类。[结果]本次研究共纳入"上火"和健康人群各35例。肠道菌群Alpha多样性指数结果显示,"上火"人群的肠道菌群多样性指数略微低于健康人群。与健康人群比较,"上火"人群肠道内含量显著上调的微生物属主要是放线菌门中的厌氧管状菌属、布劳特菌属、栖粪杆菌属、颤螺菌属、瘤胃球菌属、厌氧弧菌属、考拉杆菌属和粪芽孢菌属,显著下调的微生物属主要是拟杆菌门中的B拟杆菌属和普氏菌属。[结论]"上火"发生与肠道菌群改变密切相关,变化的肠道菌群在一定程度上能反映"上火"发生的能量代谢紊乱和黏膜免疫失衡。[Objective] To explore gut microbial community of "Shanghuo"and healthy individuals and to reveal the effects of "Shanghuo"on gut microbiome. [Methods] Stool faces from"Shanghuo"and healthy individuals were collected to extract genome DNA, and Illumina MiSeq was employed to study V3-V4 region of 16 S rRNA gene. After sequencing, QIIME was used to perform raw sequences and calculate Alpha diversity, PCoA and LEfSe analysis were used to evaluate different microbial species. [Results] This study enrolled 35"Shanghuo"individuals and 35 healthy individuals. The Alpha indices of gut microbiome in "Shanghuo"individuals were non-significant lower than that in healthy individuals. Compared with healthy individuals, the significantly increased genera in "Shanghuo"individuals were mainly the phylum Firmicutes-derived genus Anaerofustis, Blautia, Faecalibacterium,Oscillospira, Ruminococcus, Anaerovibrio, Phascolarctobacterium and Coprobacillus, the significantly decreased genera were mainly the phylum Bacteroidetes-derived genus Bacteroides and Prevotella.[Conclusion] There was a strong association between "Shanghuo"and gut microbiome, and the alterations of gut microbiome may reflect imbalances of energy metabolism and mucosal immunity in"Shanghuo"individuals.
