摘  要:目的研究血栓素A2(TXA2)、转录因子激活蛋白(AP-2α)、内皮素-1(ET-1)和β-连环素(β-catenin)在子痫前期的作用和意义。方法选取该院2015年5月至2017年5月收治的子痫前期孕妇60例,其中轻度子痫前期组、早发重度子痫前期组和晚发重度子痫前期组各20例,另选取健康孕妇30例为对照组。收集受试者血清并采用酶联免疫吸附试验(ELISA)检测血清中TXA2、AP-2α、ET-1和β-catenin的表达;采用RealTime PCR法检测血清中TXA2、AP-2α、ET-1和β-catenin基因表达。结果与对照组比较,轻度子痫前期组、早发重度子痫前期组和晚发重度子痫前期组患者TXA2、AP-2α、ET-1和β-catenin血清内水平和mRNA表达明显升高,差异具有统计学意义(P<0.05)。与轻度子痫前期组比较,早发重度子痫前期组和晚发重度子痫前期组患者TXA2、AP-2α、ET-1和β-catenin血液内水平和mRNA表达明显升高,差异具有统计学意义(P<0.05)。与早发重度子痫前期组比较,晚发重度子痫前期组患者TXA2和β-catenin血清内水平和mRNA表达明显升高,AP-2α和ET-1血清内水平和mRNA表达明显下降,差异具有统计学意义(P<0.05)。轻度子痫前期组、早发重度子痫前期组和晚发重度子痫前期组患者血清内TXA2、AP-2α、ET-1和β-catenin呈显著正相关关系(P<0.05)。结论子痫前期患者体内TXA2、AP-2α、ET-1和β-catenin呈异常过表达状态,且TXA2、AP-2α、ET-1和β-catenin与早发型子痫的严重程度具有明显的相关性。Objective To investigate the effects of thromboxane A2 (TXA2),activator protein-2α (AP-2α),endothelin 1 (ET 1) and β -catenin in pre eclampsia and its significance. Methods A total of 60 pregnant women with pre eclampsia who were treated in the hospital from May 2015 to May 2017 were enrolled in the study. They were divided into mild group, early onset severe group and late onset severe group with 20 cases each,and another 30 healthy pregnant women were taken as control group. Serum was collected and the serum levels of TXA2 ,AP 2α,ET 1 and β -catenin were detected by enzyme linked immunosorbent assay (ELISA). The gene expressions of TXA2, AP 2α,ET-1 and β -catenin in serum of patients were detected by Real-Time PCR. Results Compared with the control group, the serum levels and the expression of mRNA of TXA2, AP 2α,ET 1 and β -catenin were increased in the mild group,the early onset severe group and the late onset severe group and the differences were statistically significant (P〈0.05). Compared with the mild group,the serum levels and the expression of mRNA of TXA2,AP 2α,ET 1 and β-catenin were increased in the early-onset se vere group and the late onset severe group and the differences were statistically significant (P〈0.05). Corn pared with the early onset severe group, the serum levels and the expression of mRNA of TXA2 and β -catenin were increased in the late onset severe group and the differences were statistically significant (P〈0. 05). There was a significant positive correlation between TXA2, AP 2α,ET-1 and β -catenin in patients with mild pre eclampsia,early onset severe pre eclampsia and late onset severe pre eclampsia (P〈0.05). ConclusionThe levels of TXA2,AP 2α, ET 1 and β -catenin in pressed,and TXA2, AP 2α,ET 1 and β -catenin were pre eclampsia. patients with pre eclampsia significantly correlated with were abnormally over ex the severity of early onset pre-eclampsia.
