摘 要:目的:研究隔山消水溶性部位对阿托品所致小鼠胃肠运动及胃肠激素分泌抑制的影响。方法:将实验小鼠随机分为空白组、模型组(硫酸阿托品)、阳性药组(多潘立酮组)、隔山消水溶性部位低、中、高剂量组,多潘立酮组以0. 5 mg/10 g灌胃给予多潘立酮混悬液,隔山消低、中、高组灌胃分别给予0. 18、0. 36及0. 72 g/kg隔山消水溶性部位提取物混悬液,空白组和模型组均给予生理盐水,连续给药4 d,每天2次;末次给药30 min后,除空白组外其余5组小鼠均腹腔注射硫酸阿托品建立胃肠功能障碍模型(0. 5 mg/kg),取血、取胃及小肠组织,采用酚红测定各组胃排空率、小肠推进率,同时检测血清中胃动素(MTL)、胃泌素(GAS)、血管活性肠肽(VIP)的含量。结果:与模型组相比,隔山消水溶性部位低、中、高剂量组胃排空率和小肠推进率均显著提高(P<0. 01),其中中剂量组与多潘立酮组相当;胃肠激素水平测定结果表明隔山消水溶性部位中、高剂量组显著提高了模型小鼠的MTL、GAS含量(P <0. 05或P <0. 01),降低了VIP含量(P <0. 05)。结论:隔山消水溶性部位提取物能促进小鼠胃排空和肠推进,其机制可能与促进胃肠道MTL及GAS的释放和抑制VIP的释放有关。Objective:To study the effects of water-soluble fraction of Cynanchum wilfordii on atropine-induced gastrointestinal motility and gastrointestinal hormones in mice. Methods: The mice were randomly divided into the blank group, the model group, the domperidone group and three groups of water-soluble fraction of Cynanchum wilfordii with low dose, medium dose and high dose (0.18, 0.36 and 0.72 g/kg) . In the inhibiting state caused by atropine, the content of phenol red was determined to measure the rate of gastric emptying and small intestinal propulsion and serum levels of the motilin (MTL), gastrin (GAS) and vasoactive intestinal peptide (VIP) were measured in mice. Results: Compared with the blank group, the rates of gastric emptying and small intestinal propulsion significantly increased in the low, medium and high dose groups ( P 〈0.01), and the medium dose group was equivalent to the positive one. The MTL and GAS significantly increased ( P 〈0.05 or P 〈0.01) and VIP decreased significantly ( P 〈0.05) in the medium and high dose groups. Conclusions: Water-soluble fraction of Cynanchum wilfordii can promote the gastric emptying and small intestinal propulsion, and its mechanism is likely to be related with the promotion of MTL and GAS levels and the release of VIP level in gastrointestinal tract.
