Effects of Huoxue Tongluo Decoction on Expressions of Notch2/DLL1/Hes1 in Rabbit Modeling of SANFH

摘  要:目的研究活血通络汤在激素性股骨头缺血性坏死(SANFH)兔造模过程中Notch2/Delta-like 1 (DLL1)/发状分裂相关增强子1(Hes1)表达的影响。方法将192只日本大耳兔随机分为A、B、C、D 4组,每组各48只。B、C、D组运用贺西京造模法造模。D组第2周开始喂服中药饲料,C组第3周开始喂服中药饲料,A、B组均喂服等量普通饲料。分别于第2、5、8周末抽血检测血中的Notch2、DLL1、Hes1基因表达值,同时做股骨头病理镜检,并计算空骨陷窝率。结果第2周时,与A组比较,B组Notch2、DLL1、Hes1基因表达均升高(P<0.01);与B组比较,C组Notch2基因表达值降低(P<0.05)。与C组比较,D组Notch2基因表达值升高(P<0.01)。第5周时,与A组比较,B组Notch2、Hes1基因表达升高(P<0.01)。与C组比较,D组Notch2基因表达下降(P<0.01)。第8周时,与A组比较,B组Notch2、Hes1基因表达升高(P<0.01);与B组比较,C、D组Notch2基因表达下降(P<0.05)。空骨陷窝率:与A组比较,B组2、5、8周末空骨陷窝率明显升高(P<0.01),C、D组空骨陷窝率明显低于B组(P<0.01)。镜检结果:A组骨组织形态完整、结构清晰;B组骨小梁变细稀薄、结构紊乱、部分断裂、塌陷,脂肪细胞增多,髓腔内造血细胞减少;C组骨小梁较粗大,未见明显裂痕,出现新生成骨细胞,髓腔内造血细胞有所减少,脂肪细胞未见大量增生;D组骨小梁较为粗大、规则致密,见新生成骨细胞,髓腔内造血细胞较多。结论活血通络汤在早、中期SANFH家兔造模过程中通过调整Notch信号通路的表达从而减低SANFH的发生率,其机制可能与抑制Notch2/DLL1/Hes1信号途径的活化有关。Objective To study the effects of Huoxue Tongluo Decoction (HTD) on the expres- sions of Notch2/Delta-like 1 (DLL1)/Hesl in rabbits during the process of steroid-induced avascular necrosis of the femoral head (SANFH) modeling. Methods A total of 192 Japanese big ear rabbits were randomly divided into four groups (A, B, C and D), 48 in each. Modeling was prepared in rabbits of Group B, C, and D. Rabbits in Group D started to feed Chinese medicine forage from the second week. Those in Group C started to take Chinese medicine forage from the third week. Those in Group A and B were fed with the same amount of ordinary forage. The expressions of Notch2, DLL1, and Hesl in blood were measured at the weekend of the 2 nd, 5th, and 8th week, respectively. Meanwhile, the femoral head was pathologically examined. And the ratio of empty lacuna was calculated. Results At the 2nd week, compared with Group A, the expressions of Notch2, DLL1 and Hesl gene in Group B were all increased (P 〈0.01 ). Compared with Group B, the expression of Notch2 gene in Group C decreased (P 〈0.05). Compared with group C, Notch2 gene expression increased in Group D (P 〈0.01 ). At the 5nd week,compared with Group A, the expressions of Notch2 and Hes1 gene in Group B increased (P 〈0.01 ). Compared with group C, Notch2 gene expression decreased in Group D (P 〈0.01 ). At the 8th week, compared with Group A, the expressions of Notch2 and Hesl gene in Group B increased (P 〈0.01 ). Compared with Group B, the expression of Notch2 gene in Group C and Group D decreased (P 〈0.05). In terms of ratio of empty lacuna: Compared with Group A,the ratio of empty lacuna in Group B increased significantly at the end of the 2nd, 5th, and 8th week (P 〈0.01 ). The ratio of empty lacuna in Group D and C was obviously lower than that in Group B (P 〈0.01 ).
