不同血细胞分离机采集外周血单个核细胞的质量评估

摘　　要：目的通过比较3种型号的血细胞分离机所采集到的单采白细胞样品,及经处理后得到的外周血单个核细胞（PBMC）的质量,为选择最佳血细胞分离机机型和采集参数提供实验依据。方法分别使用,3种不同品牌的血细胞分离机Fenwal Amicus、Spectra optia、MCS~＋9000采集了20人次的外周血白细胞细胞。单采结束后对样品进行血细胞计数检测,并分离其中的外周血单个核细胞（PBMC）,染色后通过流式细胞仪进行细胞分类检测。结果 3种机型所采集到的单采白细胞样品中所含的WBC总数无统计学差异,但是Fenwal Amicus所采集的产品中混入的血小板却明显少于其他2种机型,而MCS~＋9000在采集白细胞时会混入更多的红细胞,MCS~＋9000所采集到的单采产品含有更多的中性粒细胞、嗜酸性粒细胞,而Fenwal Amicus所得到的嗜碱性粒细胞明显高于MCS~＋9000。流式细胞术分析发现,单采产品中分离出的PBMC和单核细胞绝大多数都是活细胞,主要由单核细胞和淋巴细胞组成,而粒细胞的含量很少。Baxter Amicus组最终得到的PBMC总数和T淋巴细胞总数最高,而单核细胞细胞总数则三者差异不大。结论 3种机型的血细胞分离机采集过程都比较顺利,所得到的单采白细胞样品和分离得到的PBMC质量差异不大,符合预期目的,都可用于健康志愿者的白细胞单采。Objective White blood cells（ WBCs） were collected from voluntary donors by 3 different blood cell separators and the quality of WBC samples and isolated peripheral blood mononuclear cells（ PBMCs） were compared in order to provide a support for selecting suitable type and parameters of blood cell separators. Methods 20 apheresis of white blood cells were performed with 3 different blood cell separators： Fenwal Amicus,Spectra optia and MCS~＋9000. The blood cell counts in blood samples was carried out after apheresis. PBMCs were isolated from apheresis samples and the amount,ratio and viability of different cells in PBMCs were detected by FACS. Results There was no significant difference in the number of WBCs and lymphocytes among the 3 groups of products collected by 3 different separators. Platelets in Fenwal Amicus products were significantly less than those of the other two products. MCS~＋9000 products contaminated more red blood cells,neutrophils and eosinophils,while basophils in Fenwal amicus products were significantly higher than those of MCS~＋9000 products.FACS results showed that most of isolated PBMCs and monocytes were living cells. PBMCs mainly composed of monocytes and lymphocytes as well as a small amount of granulocyte cells. The total number of PBMC and T lymphocytes were the highestin Baxter Amicus group,while the total number of mononuclear cells had no significant difference among Fenwal Amicus,Spectra optia and MCS~＋9000 groups. Conclusion This study suggested that the apheresis process of all three separators were relatively successful and the quality of apheresis WBC samples and isolated PBMCs have no huge difference,indicating that these types of cell separators can be used for WBC apheresis from healthy voluntary donors.
