摘  要:目的比较研究赤芍、白芍及其有效成分芍药苷、芍药内酯苷对原发性痛经模型小鼠的影响。方法以痛经宝为阳性对照药,采用己烯雌酚和缩宫素制备原发性痛经小鼠模型,实验一给予赤芍水提物、白芍水提物,记录潜伏期和20分钟内扭体次数。实验二采用同一造模方法给予芍药苷和芍药内酯苷进行实验,记录小鼠潜伏期和20分钟内扭体次数,并检测子宫组织中相关炎性因子水平。结果与模型组比较,白芍水提物组和赤芍水提物能使原发性痛经模型小鼠潜伏期延长、扭体次数减少,且与1 g/kg赤芍同剂量组比较,白芍水提物组的扭体次数减少(P <0. 05)。与模型组比较,芍药苷组和芍药内酯苷能使小鼠潜伏期延长、扭体次数减少,且与50 mg/kg芍药苷同剂量组比较,芍药内酯苷组的扭体次数减少(P <0. 05)。与模型组比较,100 mg/kg芍药苷组子宫组织PGF_(2α)降低、NO升高(P <0. 05); 100 mg/kg芍药内酯苷组PGF_(2α)、Ca^(2+)降低,NO升高(P <0. 01,P <0. 05,P <0. 01); 50 mg/kg芍药内酯苷组PGF_(2α)水平降低(P <0. 05)。结论在原发性痛经模型小鼠中,白芍的解痉镇痛作用略优于赤芍,芍药苷是白芍与赤芍具有解痉镇痛作用的共同有效成分,而芍药内酯苷是白芍解痉镇痛作用优于赤芍的物质基础。Objective To investigate the effects of Radix Paeoniae Rubra , Radix Paeoniae Alba and their active constituents paeoniflorin, albiflorin on spasmolysis and analgesic in mice model of primary dysmenorrhea. Methods Tongjingbao was taken as a positive control drug. The mice model of primary dysmenorrhea was established with diethylstilbestrol and oxytocin. The mice was intragastric administered to the extracts of Radix Paeoniae Rubra and Radix Paeoniae Alba , then the latency and writhing times in 20 minutes were recorded. The other experiment used the same modeling method but replaced the extracts of Radix Paeoniae Rubra and Radix Paeoniae Alba with paeoniflorin and albiflorin. Results Compared with model group, the extracts of Radix Paeoniae Alba and Radix Paeoniae Rubra could prolong latent phase and reduce the writhing times of primary dysmenorrhea model mice ( P 〈0.05). Compared with the mice which were administered 1 g/kg the extracts of Radix Paeoniae Rubra , the writhing times of the mice which were administered the same doze of the extracts of Radix Paeoniae Alba were obviously decreased ( P 〈0.05). Compared with model group, paeoniflorin and albiflorin could prolong latent phase and reduce the writhing times of primary dysmenorrhea model mice. Compared with the mice which were administered 50 mg/kg paeoniflorin, the writhing times of mice which were administered the same doze of albiflorin were obviously decreased ( P 〈0.05). Compared with model group, 100 mg/kg paeoniflorin in mice reduced the content of PGF 2 in uterine tissue and increased the content of NO ( P 〈0.05); 100 mg/kg albiflorin in mice reduced the content of PGF 2 , and Ca 2+ in uterine tissue, and increased the content of NO ( P 〈0.01, P 〈0.05, P 〈0.01 ), and 50 mg/kg albiflorin in mice reduced the content of PGF 2α in uterine tissue ( P 〈0.05). Conclusion In this model of primary dysmenorrhea mice, the analgesic effect of Radix Paeoniae Alba is better than Radix Paeoniae Rubra . Paeoniflorin
